Serial Cryomicrotomy of Saccharomyces cerevisiae for Serial Electron Cryotomography
Electron cryotomography (cryo-ET) is an increasingly popular technique to study cellular structures and macromolecules in situ. Due to poor penetration of electrons through thick biological samples, the vitreously frozen samples for cryo-ET need to be ...

An oxo–hydroxyl radical coupling mechanism is identified for O─O bond formation in Photosystem II within the low‐oxidation‐state paradigm. Ca‐bound W3 forms Ox in the S4 state and subsequently couples with O5. The resulting hydroperoxide intermediate provides a mechanistic explanation for the poor resolution of Ox in XFEL structures.

Investigating the Cytoskeleton of DRGs Using Cryo‐Electron Microscopy and Deep Learning
ABSTRACT We describe a method for determining the ultrastructural organization of axons and varicosities of cultured dorsal root ganglion (DRG) neurons using cryogenic electron microscopy (cryo‐EM). Cryo‐EM reveals the dimensions, proximity, and overall organization of biological specimens in a near‐native state, avoiding artifacts of fixation and ...

Correlative cryo-electron microscopy reveals the structure of TNTs in neuronal cells
The architecture of functional TNTs is still under debate. Here, the authors combine correlative FIB-SEM, light- and cryo-electron microscopy approaches to elucidate the structure of TNTs in neuronal cells, showing that they form structures that are ...

Preparing samples from whole cells using focused-ion-beam milling for cryo-electron tomography
Recent advances have made cryogenic (cryo) electron microscopy a key technique to achieve near-atomic-resolution structures of biochemically isolated macromolecular complexes.

In situ microfluidic cryofixation for cryo Focused Ion Beam milling and cryo electron tomography. [PDF]
We present a microfluidic platform for studying structure-function relationships at the cellular level by connecting video rate live cell imaging with in situ microfluidic cryofixation and cryo-electron tomography of near natively preserved, unstained ...

Anatomy of a SPAN Particle: A Four‐Level Structural Framework for Lithium–Sulfur Batteries
Sulfurized polyacrylonitrile (SPAN) is a key cathode material for batteries. Borrowing the four‐level classification of protein structure, this perspective demonstrates SPAN's structural complexity into well‐defined levels, giving researchers clear research boundaries and targets at each level for improving battery performance.

In situ high-resolution cryo-EM reconstructions from CEMOVIS
Cryo-electron microscopy can be used to image cells and tissue at high resolution. To ensure electron transparency, the sample thickness must not exceed 500 nm.
