Switching on Supramolecular DNA Junction Binding Using a Human Enzyme

open access: yesAngewandte Chemie, EarlyView.
Stimulus responsive control of DNA junction formation: Enzyme action cleaves flexible arms from a metallo‐supramolecular cylinder, transforming an inert metallo‐supramolecular structure into an active metallo‐supramolecular agent, and switching on its ability to assemble, bind and stabilize DNA three‐way junction structures.

An Artificial Metal‐Free Peroxidase Designed Using a Ferritin Cage for Bioinspired Catalysis

open access: yesAngewandte Chemie, EarlyView.
Designing artificial enzymes remains challenging due to the need for precise active site control. Here, we present a simple strategy to create metal‐free artificial enzymes by engineering Histidine clusters inside a ferritin cage. The resulting catalyst mimics peroxidase activity, and its stable cage structure offers new insights into supramolecular ...

N‐Insertion of Diazonium Salts Into Ketone Derivatives

open access: yesAngewandte Chemie, EarlyView.
Diazonium salts insert into ketones and their silyl enol ethers through a sequence of α‐azenylation and ring expansion via an N‐iminoaziridinium species. Coupled with an efficient N‐N bond cleavage, this protocol offers a mild alternative to Beckmann rearrangements and a powerful tool for skeletal editing of ketones.

Cooperativity of Electron Transfer Coupled Spin Transitions in a Tetranuclear Fe/Co Prussian Blue Analogue Revealed by Ultrafast Spectroscopy

open access: yesAngewandte Chemie, EarlyView.
Ultrafast pump‐probe spectroscopies show that irradiation of a square Fe/Co Prussian blue analogue induces electron transfer within one Fe–Co subunit followed by fast spin crossover, and coupled equilibria between all states trigger a second electron transfer coupled to spin crossover in the other Fe–Co subunit on a ns timescale.

Structural Basis for the Catalytic Mechanism of ATP‐Dependent Diazotase CmaA6

open access: yesAngewandte Chemie, EarlyView.
Several diazotases have been reported in recent years, but the detailed catalytic mechanism is unclear. From X‐ray crystallography and Cryo‐EM analysis, we obtained the structure of a diazotase CmaA6, which synthesizes 3‐diazocoumaric acid. Kinetic analysis and computational analysis indicated that CmaA6 catalyzes diazotization via an unprecedented ...

Isomerisation and Insertion Chemistry of Imidosilanes Enabled by Reversible Si(IV)/Si(II) Redox Shuttling

open access: yesAngewandte Chemie, EarlyView.
We report rearrangement and functionalisation chemistry at silicon facilitated by reductive ligand migration and Si(II)/Si(IV) redox shuttling. Abstract Spontaneous redox shuttling at silicon is very rare, primarily reflecting the thermodynamic challenges associated with reduction processes for lighter p‐block elements.

Enhancing Cold Adaptation of Bidomain Amylases by High‐Throughput Computational Engineering

open access: yesAngewandte Chemie, EarlyView.
This work uncovers molecular determinants of cold adaptation in bidomain amylases by combining MD simulations and biochemical assays, revealing that increased domain separation drives enhanced activity at low temperatures. Guided by this principle, high‐throughput in silico linker screening of mesophilic psA was performed, yielding variant psA121 with ...

Iridium‐Catalyzed Asymmetric Allenylic Substitution via Kinetic Resolution Enabled by New Monodentate Ligands

open access: yesAngewandte Chemie, EarlyView.
The first kinetic resolution of allenylic alcohols by asymmetric allenylic substitution has been achieved, which was enabled by the design of a new chiral ligand that altered the energetic profiles of the previously established pathways. Both secondary and tertiary alcohols were suitable substrates.
