Cutaneous squamous cell carcinoma
The squamous cell carcinoma (SCC) is the most frequent cutaneous carcinoma, after basal cell carcinomas. Cutaneous SCC have a good prognosis, in most cases, but the achievement of elderly people and the frequent recurrence, made a real health problem. CSPG4 is identified as a high‐value, stemness‐associated target in HPV‐negative HNSCC. By implementing rational biophysical engineering, a humanized and charge‐optimized CAR is developed to overcome tonic signaling‐induced exhaustion. This strategy induces a profound transcriptomic shift toward a rejuvenated, stem‐like memory state, significantly ...

This review surveys nanoparticle‐based strategies to enhance adoptive cell therapy, particularly CAR‐T cell approaches, in solid tumor treatment. It describes how nanoparticles can improve tumor immunogenicity and T‐cell infiltration while reducing toxicity, and how they enable in vivo CAR‐T cell generation.
