Interpreting the effects of DNA polymerase variants at the structural level

open access: yesMolecular Oncology, EarlyView.
Using MAVISp and molecular dynamics simulations, we analyzed over 60 000 missense variants in POLE and POLD1 from ClinVar, COSMIC, cBioPortal, and saturation mutagenesis. Identified mechanistic indicators, including stability, binding, and long‐range, enable structural interpretation, providing ACMG‐like evidence for possible reclassification of VUS ...

A Finitely Converging Cutting Plane Technique [PDF]

open access: yes, 1984
We consider a new finitely convergent cutting plane algorithm for mixed integer linear programs in which the optimal objective value is assumed to be integral.

Proximal Analytic Center Cutting Plane Algorithms for Variational Inequalities and Nash Economic Equilibrium

open access: yesMathematics
In this study, we proposed proximal analytic center cutting plane algorithms for solving variational inequalities whose domains are normal regions. Our algorithms stop with a solution of the variational inequality after a finite number of iterations, or ...

Metastasis on pause: How dormant tumor cells stay hidden within the tumor microenvironment and evade immune surveillance

open access: yesMolecular Oncology, EarlyView.
Dormant cancer cells can hide in distant organs for years, evading treatment and the immune system. This review highlights how signals from the surrounding tissue and immune environment keep these cells inactive or trigger their reawakening. Understanding these mechanisms may help develop therapies to eliminate or control dormant cells and prevent ...

Developmental programmes drive cellular plasticity, disease progression and therapy resistance in lung adenocarcinoma

open access: yesMolecular Oncology, EarlyView.
This study shows that lung adenocarcinomas exploit developmental branching morphogenesis to acquire a therapy resistant basal‐like tumour cell state. This process was found to be regulated by combined TP53 loss‐of‐function and type‐I interferon signalling, identifying a novel axis for biomarker and therapeutic target discovery.

Stimulator of interferon genes agonist augmented antitumor immunity of osimertinib in Egfr‐mutated lung cancer

open access: yesMolecular Oncology, EarlyView.
Combining osimertinib with the STING agonist ADU‐S100 activates innate and adaptive immunity to overcome the non‐inflamed microenvironment of Egfr‐mutant lung cancer. This combination increases NK and CD8+ T‐cell infiltration, associated with activation of the STING‐IRF3 pathway and local immunogenic cell death.

An Efficient Cutting Plane Algorithm for Unconstrained Convex Minimization [PDF]

open access: yes, 1979
A cutting-plane algorithm is proposed for minimizing a continuously differentiable convex function of n variables. One notable feature of this algorithm is that each cut is actually a supporting hyperplane, which does not require an iterative procedure ...

Cutting Plane Algorithms for Semidefinite Relaxations [PDF]

open access: yes, 1997
We investigate the potential and limits of interior point based cutting plane algorithms for semidefinite relaxations on basis of implementations for max-cut and quadratic 0-1 knapsack problems.
