Finding novel vulnerabilities of hypomorphic BRCA1 alleles

open access: yesMolecular Oncology, EarlyView.
Synthetic lethality screens performed to identify novel vulnerabilities often model complete gene loss, thereby overlooking patient‐derived hypomorphic mutations. In this study, we have performed genome‐wide CRISPR screens on BRCA1 hypomorphic mutations, showing BRCA1I26A behaves like wild‐type, while BRCA1R1699Q mimics deficiency. Furthermore, we have

MITF maintains genome stability in nonmelanocyte lineages

open access: yesMolecular Oncology, EarlyView.
MITF is essential for melanocyte survival and acts as an oncogene in 10%–20% of melanomas. We show that MITF depletion causes genome instability in nonmelanocytic cells, leading to LATS2‐mediated P53 activation, cell cycle arrest, and apoptosis. This study highlights the role of MITF as a genome maintenance factor beyond the melanocyte lineage. Created

The psychology of multimedia databases

open access: yes, 2000
Multimedia information retrieval in digital libraries is a difficult task for computers in general. Humans on the other hand are experts in perception, concept representation, knowledge organization and memory retrieval.

Exploring Comprehension Strategies of Modular Process Models: A Combined Eye-Tracking and Concurrent Think-Aloud Study

open access: yesBrain Sciences
The study of complex process models often encounters challenges in terms of comprehensibility. This paper explores using modularization as a strategy to mitigate such challenges, notably the reduction in complexity.

A novel quinazolinone insulin receptor inhibitor and its synergy with an EGFR inhibitor in glucose‐driven glioblastoma

open access: yesMolecular Oncology, EarlyView.
The novel styrylquinazolinone‐based molecule W1B effectively suppresses glioblastoma by inhibiting IGF1R and EGFR. In high‐glucose microenvironments driving tumor resistance, W1B acts synergistically with the EGFR inhibitor dacomitinib. This combination safely blocks compensatory survival signaling in zebrafish xenograft models. Showcasing promising in

On the unification of active databases and deductive databases [PDF]

open access: yes, 1993
These two rule-oriented paradigms of databases have been the focus of extensive research and are now coming of age in the commercial DBMS world. However, the systems developed so far support well only one of the two paradigms—thus limiting the effectiveness of such systems in many applications that require complete integration of both kinds of rules ...

Optimizing Spatial Databases [PDF]

open access: yes
This paper describes the best way to improve the optimization of spatial databases: through spatial indexes. The most commune and utilized spatial indexes are R-tree and Quadtree and they are presented, analyzed and compared in this paper. Also there are

Oncogenic DMTF1β promotes cancer cell motility by regulating autophagy through ULK1 stabilization

open access: yesMolecular Oncology, EarlyView.
In the current study, we demonstrate that the oncogene DMTF1β regulates ULK1 stability by reducing its proteasomal degradation in cancer cells. This stabilization enables ULK1 to induce autophagy, which in turn facilitates cancer cell migration. Consequently, reduced DMTF1β levels lead to decreased autophagy and impaired cancer cell migration.

Liquid biopsy‐based diagnostic evaluation of hypermethylated CpG sites for ovarian cancer diagnosis

open access: yesMolecular Oncology, EarlyView.
This schematic outlines the workflow from biomarker identification to duplex MethyLight assay validation for epithelial ovarian cancer diagnosis using cfDNA‐based liquid biopsy. Initial screening of hypermethylated CpG candidates (cg02957270, cg10061138 cg00480298, COL2A1) was performed in tissue using ARMS‐PCR, COBRA, qPCR and image analysis. Selected
