This study shows that lower NAM levels in PE‐derived pEVs correlate with disease severity. NAM‐deficient pEVs reduce Th1 and Th17 inhibition, leading to PE‐like symptoms. NAM in pEVs inhibits Th1 via SIRT1 and Th17 via macrophages. Reduced NAM in PE‐EVs is due to decreased HRS expression in trophoblasts, resulting from elevated HSP27.

The placenta is a unique organ with various immunological and endocrinological roles that modulate maternal and fetal physiology to promote maternal-fetal tolerance, pregnancy maintenance, and parturition at term. During pregnancy, the hormone prolactin (

The interaction between trophoblasts and decidual polymorphonuclear myeloid‐derived suppressor cells (dPMN‐MDSCs) via the XCL1–XCR1 axis is crucial for fetal development during the third trimester. Disruption of this axis impairs FOXO1 activity and causes metabolic imbalance in dPMN‐MDSCs, contributing to adverse outcomes associated with advanced ...

T lymphocytes in the third trimester decidua in preeclampsia
Objective: The aim of study was to conduct immunohistochemical quantification of CD3+ and CD8+ decidual lymphocytes in preeclampsia. Methods: A study group included 30 cases of preeclampsia and a control group included 20 healthy pregnant women, all ...

Expression of Estrogen Receptor α by Decidual Macrophages in Preeclampsia
Preeclampsia is a gestation-associated hypertensive syndrome that threatens the life and health of the mother and the child. The condition is presumably caused by systemic failure with a strong involvement of innate immunity.

In trophoblast cells, T/S (TNFα + SM164) activated caspase‐3, which cleaved GSDME to switch apoptosis to pyroptosis, causing cell swelling, membrane rupture, and release of LDH, HMGB1, IL‐1β and IL‐18. These cytokines induced pro‐inflammatory macrophage polarization, which in turn reinforced pyroptotic signaling in trophoblasts, amplifying systemic ...

Innate Immune Mechanisms to Protect Against Infection at the Human Decidual-Placental Interface
During pregnancy, the placenta forms the anatomical barrier between the mother and developing fetus. Infectious agents can potentially breach the placental barrier resulting in pathogenic transmission from mother to fetus.
