UCHL3 Regulates Subgenomic Flaviviral RNA Condensates to Promote Virus Propagation
ABSTRACT Flavivirus subgenomic RNAs (sfRNAs) antagonise antiviral defences, yet how sfRNAs are organized and maintained in cells remains poorly understood. Here we identify ubiquitin C‐terminal hydrolase L3 (UCHL3) as a post‐translational regulator of flavivirus sfRNA stability and function.

Host‐specific compatibility between RNF138‐like proteins and flavivirus NS5 determines NS5 stability. Mammalian RNF138 but not arthropod homologs recognizes and induces conserved NS5/RdRp K48‐linked ubiquitination and proteasomal degradation, thereby restricting viral replication. Upon JEV infection, ZNF33B recruits METTL14 to stabilize the METTL3‐METTL14 m6A methyltransferase complex, leading to increased m6A modification of host transcripts, including Trim25 mRNA. ZNF33B selectively binds m6A‐modified sites on Trim25 mRNA and accelerates its decay, resulting in reduced TRIM25 protein abundance.

Dual Blockade of LILRB1 and LILRB2 Enhances Antiviral Immune Responses in SIV Infection
Dual LILRB1/B2 blockade with mac20G10 reshapes myeloid activation during acute SIV infection by targeting LILRB1 and LILRB2 on myeloid cells. This treatment enhances CD80 expression on selected myeloid subsets and increases plasma IFN‐λ, IL‐8, and IL‐1RA.

An on‐resin dual‐modification strategy for peptides is presented, comprising S‐alkynylation of peptide thiosulfonates and regioselective iridium‐catalyzed azide–thioalkyne cycloaddition. This method reliably provides access to peptides with intricate non‐canonical modifications, being compatible with complex peptides, alkynes and azides.
