A targeted metabologenomic strategy integrating genomic and metabolomic hallmarks enabled the discovery of two new thioamides, thiogochangamides A and B, from bacteria. Their absolute configurations, previously unresolved within the thioviridamide family, were fully assigned through various chemical derivatizations and computational analysis ...

Delivery of Pleckstrin‐Homology Domains Suppresses PI3K/Akt Signaling and Breast Cancer Metastasis
Current therapies curb tumor growth but not metastasis. Obscurin, a giant metastasis suppressor lost in breast cancer, restrains PI3K/Akt signaling but is impractical to restore. We deploy a mini‐obscurin, comprising the obscurin‐PH‐domain, which sequesters PI3K‐p85, potently suppressing invasion and metastasis.

Comparison of clinical and genetic characteristics between Dent disease 1 and Dent disease 2
Background Dent disease is associated with low molecular weight proteinuria and hypercalciuria and caused by pathogenic variants in either of two genes: CLCN5(Dent disease 1) and OCRL(Dent disease 2). It is generally not accompanied by extrarenal manifestations and it is difficult to distinguish Dent disease 1 from Dent disease 2 without gene testing ...

A Focus on the Metabolic Dysfunction of Proximal Tubule Cells in Dent Disease Type 1
Dent disease type 1 is a rare inherited renal disorder affecting mainly young males, generally leading to end-stage renal failure and for which there is no cure. It is caused by inactivating mutations in the gene encoding ClC-5, a 2Cl-/H+ exchanger found on endosomes in the renal proximal tubule.

Sex‐Specific Regulation of Glycemic Homeostasis by Theabrownin from Pu‐erh Tea
Pu‐erh tea's key component, theabrownin (TB), lowers blood glucose in a sex‐specific manner. In females, estrogen boosts intestinal MUC2 production, which dramatically enhances TB's ability to inhibit the carbohydrate‐digesting enzyme α‐glucosidase.

Model of circNrip1 (cNrip1) upregulation driving neuropathic pain mechanisms. After peripheral nerve injury, increased FUS triggers the formation and upregulation of cNrip1 in injured DRG neurons. Upregulated cNrip1 recruits SYNCRIP to the 3′‐UTR of Tlr2 mRNA by binding to both, thereby promoting SYNCRIP‐triggered Tlr2 mRNA stability and increasing ...

ABSTRACT Base editors enable precise genome modification and have emerged as a promising therapeutic approach for correcting diseases caused by single‐nucleotide variants. While the current efficient version of adenine base editors (ABEs), such as ABE8e, exhibits exceptional efficiency for A‐to‐G conversions, their clinical translation is hindered by ...
