In the title compound, 2-amino-7-(2-deoxy-beta-D-erythro-pentofuranosyl)-3,7-dihydropyrrolo[2,3-d]pyrimidin-4-one, C11H14N4O4, the N-glycosylic bond torsion angle, chi, is anti [-106.5 (3) degrees ]. The 2'-deoxyribofuranosyl moiety adopts the 3T4 (N-type) conformation, with P = 39.1 degrees and tau(m) = 40.3 degrees .

Deoxyguanosine kinase (ATP:deoxyguanosine 5'-phosphotransferase) has been purified up to a specific activity of 10.3 nmol/min per mg protein from human placenta. The enzyme appears to have a molecular weight of 58 000 from the results of Sephadex G-75 gel filtration.

The metal ions Cu(II), Fe(II), and Cr(III) were allowed to react with H(2)O(2) in the presence of either the mononucleoside 2'-deoxyguanosine (dG) or the mononucleotide 2'-deoxyguanosine-5'-monophosphate (dGMP). The percentage of reacted dG or dGMP that formed the oxidative damage marker 8-hydroxy-2'-deoxyguanosine (8-OH-dG) was monitored.

Structure of a neothramycin-2′-deoxyguanosine adduct

Abstract The interaction of neothramycin, a pyrrolo (1,4)benzodiazepine antibiotic with antitumor activity, with deoxyribonucleosides was studied as a model of the drug-DNA binding. The antibiotic was demonstrated to react with 2′-deoxyguanosine in aqueous dimethyl sulfoxide. Two reaction products were found on thin-layer plates.

Hydroxylation of Deoxyguanosine in DNA by Copper and Thiols

DNA was incubated with glutathione (GSH) and copper and then assayed for 8-hydroxydeoxyguanosine (8-OHdG) in order to better understand the antioxidant and prooxidant characteristics of GSH in copper-dependent DNA damage. Ratios of GSH to Cu(II) less than 3 resulted in 8-OHdG production; however, higher ratios did not generate 8-OHdG.
