Phosphorylation of Deoxyguanosine in Rat Liver Mitochondria

Recent studies document that mitochondria from mouse skin (1), human lung fibroblasts (2) and calf thymus (3) contain a deoxyguanosine (dG) kinase which is distinctly different from that found in the cytosol. A preliminary report (4) has shown that dG is taken up by intact, respiring rat liver mitochondria by a carrier mediated process.

The reaction of melphalan with deoxyguanosine and deoxyguanylic acid

The reaction of melphalan (phenylalanine mustard, I) with 2'-deoxyguanosine, followed by removal of the sugar in acid, yielded two products. The major product was identified as 4-(N-(2-guanin-7-ylethyl)-N-(2-hydroxyethyl)amino)phenyl- alanine (II) by ultra-violet absorption, mass and NMR spectroscopy.

8-Hydroxy-2′-deoxyguanosine formation during the catalytic oxidation of hydrazines in the presence of 2′-deoxyguanosine

The reaction of 2'-deoxyguanosine (1), substituted hydrazines (Hy) and oxygen, in the presence of Cu(II) as catalyst, yields 8-hydroxy-2'-deoxyguanosine (2). The rate of formation of 2 was found to be dependent upon the oxidation potential of the Hy and on structural factors of the Hy.

The reactivity of peroxynitrite (OONO-) with DNA is of interest because it is is released during chronic inflammation, a major contributor to cancer. Peroxynitrite was found to undergo homolytic addition to the C4-C5 double bond of 2'-deoxyguanosine with formation of 4,5-dihydro-5-hydroxy-4-(nitrosooxy)-2'-deoxyguanosine (nox-dG).

AbstractBackground:Hepatic involvement is a common feature in childhood mitochondrial disorders. Deoxyguanosine kinase (DGUOK) deficiency is one of the mitochondrial DNA depletion syndromes associated with hepatocerebral syndrome. Hepatic disease and neurologic dysfunction occurs within weeks after birth. Low birth weight is one of the common features.
