DNA methyltransferase inhibitors in hematological malignancies and solid tumors
Abstract Epigenetic modifications such as DNA methylation play a fundamental role in oncogenesis and the progression of neoplasms neoplasias. DNA methyltransferase inhibitors (DNMTi) constitute a family of therapeutic agents that impede the methylation at the 5‐position on cytosine nucleotides, thereby modulating the epigenetic regulation of tumor ...

Al‐Hilfi et al. present a biocatalytic strategy for synthesizing 5‐methyl‐5,6‐dihydrothymidine (5‐MDHT), a sensitive MRI contrast agent. The study demonstrates that recombinant enzyme catalysis offers an efficient, sustainable, and eco‐friendly alternative to traditional chemical synthesis for producing clinically relevant imaging probes.

Further investigation of the synthesis of deoxyribonucleoside-3' (t-butyl) O-(2-cyanoethyl) phosphorothioites as monomer building units for the phosphorothioite approach has led us to conclude that internucleotidic bond formation proceeded via bis(deoxyribonucleoside-3') (2-cyanoethyl) phosphite intermediates, which proved to be activated by iodine ...
