Synthesis of Ser-Ala-Ala Cyclic Desmosine

Synthesis, 2023
AbstractElastin is present in the extracellular matrix of various tissues, most abundantly in the aorta and major vascular vessels, and is formed by self-assembly followed by concomitant crosslinkers of tropoelastin, an elastin precursor. Desmosine is a pyridinium-based tetrafunctional amino acid that serves as an important crosslinker to bind the ...

Synthesis of Multi‐Deuterated Desmosine

ChemistrySelect, 2020
Abstract Desmosine, a crosslinking pyridinium amino acid found in elastin, is a useful biomarker for the diagnosis of diseases related to elastin degradation, such as chronic obstructive pulmonary disease (COPD).

The biosynthesis of desmosine and isodesmosine II. Incorporation of specifically labeled lysine into desmosine and isodesmosine

Biochimica et Biophysica Acta (BBA) - Protein Structure, 1967
Abstract 1. 1.|The incorporation of radioactivity into desmosine and isodesmosine from [I- 14 C]lysine and [6- 14 C]lysine was studied using chick embryo aortae in tissue culture. 2. 2.|The side chain attached to the pyridinium nitrogen in desmosine and iso-desmosine was liberated in the form of lysine by the action of strong alkali. 3. 3.

Pharmacokinetics of Intravenously Administered Desmosine in Sheep

Connective Tissue Research, 1984
The pharmacokinetics of desmosine following a 5 mg intravenous dose was determined in five sheep. The biological half-life (t 1/2 beta) of desmosine varied between 81-489 min. Urinary recovery of unchanged desmosine indicated that the compound is not extensively metabolized and that the primary route of elimination is via the kidney.

Desmosine Biosynthesis: Nature of Inhibition by D-Penicillamine

Science, 1968
Administration of D-penicillamine and lathyrogens such as β-amino-propionitrile to animals markedly alters connective tissue by preventing the normal cross-linkage of elastin and collagen. It had been shown that β-aminopropionitrile blocks the cross-linkage of elastin and collagen by preventing the initial step in cross-linkage: the conversion of ...

Photolysis and Ozonolysis of Desmosine and Elastolytic Peptides

Connective Tissue Research, 1985
This paper describes the combined effects of photolysis and ozonolysis upon the main interchain crosslinks of elastin, desmosine and isodesmosine. Photolysis of purified (iso)desmosines in solution leads to the cleavage of the pyridinium rings to give lysine and an analogue of glutaconic aldehyde which is deformylated to a stable compound absorbing at ...

Mature bovine descemet's membrane contains desmosine and isodesmosine

Biochemical and Biophysical Research Communications, 1982
The major cross-links of mature bovine Descemet's membrane have been compared with those of the anterior lens capsule. Both basement membranes contained hydroxylysinonorleucine but Descemet's membrane also contained two components with the chromatographic properties of desmosine and isodesmosine.

Desmosines: A rapid single-column isolation procedure

Analytical Biochemistry, 1972
Abstract A simple and rapid method is described for the isolation of the desmosines from elastin. Elastin is hydrolyzed with 6 N HCl in a conventional manner, treated with decolorizing charcoal, and adjusted to pH 2.8. Gel permeation on Bio-Gel P-2 resolves an ultraviolet-absorbing peak eluted between cytochrome c and cyanocobalamin.

Urinary excretion of desmosine in patients with severe burns

Metabolism, 1981
Urinary excretion of total desmosine was measured by a radioimmunoassay in severely burned adult males, as well as in normal adult males. Total urinary desmosine was significantly elevated in all the samples in the burned patients, who had injuries involving more than 19% of total body surface area.

Photolysis of Desmosine And Isodesmosine by Ultraviolet Light

1977
It is known that the pyridinium ring of a model compound such as N-methyl pyridinium chloride is cleaved by U.V. radiation at 254 nm. At acid pH the products obtained are methylamine and glutaconaldehyde. Under the same conditions desmosine and isodesmosine are degraded into lysine and probably into the homologous substituted ketone of glutaconaldehyde.
