Synthesis and LC-MS/MS analysis of desmosine-CH2, a potential internal standard for the degraded elastin biomarker desmosine

Org. Biomol. Chem., 2014
Chemical synthesis and LC-MS/MS analysis of desmosine-CH2, a potential internal standard for the degraded elastin biomarker desmosine, are achieved.

[43] Antibodies to desmosine

1982
Publisher Summary With the exception of eggshell membrane, elastin is the only known protein to contain desmosine. Because desmosine levels in elastin are constant within species, this cross-linking amino acid can be used to quantitate mature, insoluble elastin. This chapter presents the procedure of radioimmunoassay specific for desmosine.

Polyclonal anti-desmosine antiserum is specific to desmosine molecule: no cross-reaction to pyridinoline.

The Tokai journal of experimental and clinical medicine, 1991
Inhibition ELISA assay was used to examine the cross-reaction of the polyclonal anti-desmosine antiserum produced in rabbit against molecules possessing a "pyridinium ring" as their core structure i.e. isodesmosine, pentasine, pyridinoline and 2'-deoxypyridinoline, highly purified with column chromatography, and structurally unrelated substances, i.e ...

Urinary desmosine as a biomarker in acute lung injury

Biomarkers, 2006
Acute lung injury (ALI) is a complex disorder associated with an acute inflammatory response thought to contribute to tissue injury. Desmosine, a cross-linking amino acid present in elastin, is released during matrix degradation and cleared by the kidney.

Fractionation of Urine to Allow Desmosine Analysis by Radioimmunoassay

Annals of Clinical Biochemistry: International Journal of Laboratory Medicine, 1992
The present study was designed to re-evaluate the radioimmunoassay for desmosine in urine, which is currently used as a measure of elastin metabolism. Using ion exchange chromatography, gel filtration and affinity chromatography it was shown that at least five other compounds in hydrolysates of human urine competed for desmosine in the RIA ...

Ion-exchange column chromatography of desmosine and isodesmosine

Analytical Biochemistry, 1969
Abstract A modification of a standard amino acid analysis procedure is used for the resolution and quantification of the polyfunctional amino acids desmosine and isodesmosine. In addition to the long and short columns normally used during an amino acid analysis, a third column (60 cm resin height) packed with Beckman Custom Research Resin type PA-28 ...

Total synthesis of COPD biomarker desmosine that crosslinks elastin

Chemical Communications, 2012
Desmosine, a crosslinking amino acid of elastin, is an attractive biomarker for diagnosis of chronic obstructive pulmonary disease (COPD). In this study, the first total synthesis of (+)-desmosine was achieved in 11% overall yield in 13 steps utilizing stepwise and regioselective Sonogashira cross-coupling reactions.

Free Desmosine is a Sensitive Marker of Smoke-Induced Emphysema

Lung, 2018
While the elastin-specific crosslinks, desmosine and isodesmosine (DID), are increased in blood, urine, and sputum of patients with clinically documented pulmonary emphysema, the usefulness of DID in detecting early lung injury remains untested. To this end, our laboratory has measured DID in a hamster model of smoke-induced emphysema, involving only ...

Desmosine radioimmunoassay for measuring elastin degradation in vivo.

The American review of respiratory disease, 1981
Desmosine is a cross-link amino acid unique to elastin. Previous work has shown that during turnover in the body, desmosine is not reused, and that desmosine is not absorbed from the intestine. Instead, all desmosine released in the course of elastin metabolism is excreted in the urine attached to low molecular weight peptides.
