High levels of desmosines in urine and plasma of patients with pseudoxanthoma elasticum

European Journal of Clinical Investigation, 2004
AbstractBackground  Pseudoxanthoma elasticum (PXE), a rare heritable disorder caused by mutations of the ABCC6 gene, is characterized by fragmentation and mineralization of elastic fibres. We determined the extent of degradation of elastin by measuring and comparing the amount of desmosines in plasma and urine of PXE patients, healthy carriers and ...

Urinary assays for desmosine and hydroxylysylpyridinoline in the detection of cirrhosis

Journal of Hepatology, 1997
Non-invasive markers of liver fibrosis have great potential for both the diagnosis and therapy of liver disease and cirrhosis. The aim of this study was to evaluate the potential of urinary amino acids desmosine (DES) and isodesmosine (IDES) derived from the breakdown of elastin and hydroxylysylpyridinoline (HP) and lysylpyridinoline (LP) derived from ...

[43] Antibodies to desmosine

1982
Publisher Summary With the exception of eggshell membrane, elastin is the only known protein to contain desmosine. Because desmosine levels in elastin are constant within species, this cross-linking amino acid can be used to quantitate mature, insoluble elastin. This chapter presents the procedure of radioimmunoassay specific for desmosine.

The presence in elastin of possible cyclic precursors of desmosine and isodesmosine

Biochemical and Biophysical Research Communications, 1971
Abstract Mass spectral evidence is presented that the elastin of bovine ligamentum nuchae contains, in addition to desmosine and isodesmosine, larger levels of their respective cyclic dehydrodesmopiperidine precursors. In addition to being precursors of the desmosines, such compounds are most likely cross-links in themselves.

Ion-exchange column chromatography of desmosine and isodesmosine

Analytical Biochemistry, 1969
Abstract A modification of a standard amino acid analysis procedure is used for the resolution and quantification of the polyfunctional amino acids desmosine and isodesmosine. In addition to the long and short columns normally used during an amino acid analysis, a third column (60 cm resin height) packed with Beckman Custom Research Resin type PA-28 ...

A Large Scale Procedure for Purification of Desmosine and Isodesmosine

Preparative Biochemistry, 1975
A rapid large scale procedure was devised for the purification of desmosine and isodesmosine from ligamentum nuchae elastin. The method makes use of the hydrophilic nature of the desmosines which preferentially absorbs to cellulose fibers in mixtures of organic solvents. Resolution of the isomers was achieved on a polystyrene resin column.

Desmosine Radioimmunoassay as a Means of Studying Elastogenesis in Cell Culture

Connective Tissue Research, 1981
Elastin is synthesized by fibroblasts and chondroblasts in cell culture shortly before the cells become confluent. Fibroblasts secrete elastin into the medium as soluble tropoelastin molecules, which form desmosine crosslinks and become constituents of the cell layer only after three weeks in culture.
