Publisher Summary With the exception of eggshell membrane, elastin is the only known protein to contain desmosine. Because desmosine levels in elastin are constant within species, this cross-linking amino acid can be used to quantitate mature, insoluble elastin. Biochemical and Biophysical Research Communications, 1971Abstract Mass spectral evidence is presented that the elastin of bovine ligamentum nuchae contains, in addition to desmosine and isodesmosine, larger levels of their respective cyclic dehydrodesmopiperidine precursors. In addition to being precursors of the desmosines, such compounds are most likely cross-links in themselves.

A Large Scale Procedure for Purification of Desmosine and Isodesmosine
Preparative Biochemistry, 1975A rapid large scale procedure was devised for the purification of desmosine and isodesmosine from ligamentum nuchae elastin. The method makes use of the hydrophilic nature of the desmosines which preferentially absorbs to cellulose fibers in mixtures of organic solvents. Resolution of the isomers was achieved on a polystyrene resin column.

Desmosine Radioimmunoassay as a Means of Studying Elastogenesis in Cell Culture
Connective Tissue Research, 1981Elastin is synthesized by fibroblasts and chondroblasts in cell culture shortly before the cells become confluent. Fibroblasts secrete elastin into the medium as soluble tropoelastin molecules, which form desmosine crosslinks and become constituents of the cell layer only after three weeks in culture.
