Indoleamine 2,3 dioxygenase-1 (IDO1) catalyzes tryptophan-kynurenine metabolism in many inflammatory and cancer diseases. Of note, acute inflammation that occurs immediately after heart injury is essential for neonatal cardiomyocyte proliferation and ...

Macrophages exposed to macrophage colony-stimulating factor acquire the capacity to suppress T cell proliferation; this effect is associated with de novo expression of the tryptophan-catabolizing enzyme indoleamine 2,3-dioxygenase (IDO). We have purified

Stimuli‐Responsive Supramolecular Biomaterials for Cancer Theranostics
The ultimate goal of cancer theranostics is to get imaging agents and therapeutic cargo to tumor sites when and where they are required. “Smart” systems should be developed. This review discusses the characteristics of physiological stimuli, types and action modes of external stimuli, construction approaches and working principles, as well as ...

Catalytic mechanism and molecular engineering of quinolone biosynthesis in dioxygenase AsqJ
The catalytic activity of dioxygenase AsqJ is strictly relying on the methylation of quinolone substrates. Here, the authors apply molecular simulations, X-ray crystallography and in vitro biochemical studies to the engineering of dioxygenase AsqJ with ...

Carnitine metabolism to trimethylamine by an unusual Rieske-type oxygenase from human microbiota [PDF]
Dietary intake of L-carnitine can promote cardiovascular diseases in humans through microbial production of trimethylamine (TMA) and its subsequent oxidation to trimethylamine N-oxide (TMAO) by hepatic flavin-containing monooxygenases.

Gabpα‐Pparγ Complex Determines Glycolytic Capacity and Lactic Acid Homeostasis in Brown Fat
This study identifies the Gabpα–Pparγ complex as a key transcriptional regulator that couples glycolysis to lactate‐driven thermogenesis in brown adipose tissue by directly activating Eno1 transcription. Competitive disruption of this interaction impairs lactate metabolism and thermogenic capacity.

IR‐induced dysbiosis depletes P. coprophilus and its metabolite 6‐methyluracil, leading to disinhibition of the IDO1‐Kyn‐AHR axis. This results in sustained fibroblast activation and collagen deposition, driving radiation induced intestinal fibrosis. ABSTRACT Therapeutic options for radiation‐induced intestinal fibrosis (RIF) remain limited. This study

Fluoxetine Prevents the Development of Depressive-like Behavior in a Mouse Model of Cancer Related Fatigue [PDF]
Cancer patients frequently suffer from fatigue, a complex syndrome associated with tiredness and depressed mood. Cancer-related fatigue (CRF) can be present at the time of diagnosis, escalates during treatment, and can persist for years after treatment ...
