Desensitization for drug allergy
Current Opinion in Allergy and Clinical Immunology, 2006Desensitization for drug allergy is the induction of temporary clinical unresponsiveness to drug antigens. Gradual reintroduction of small doses of drug antigen at fixed time intervals allows for the delivery of full therapeutic doses, protecting patients from anaphylaxis.

The reasons for this are not clear, but are likely to be multifactorial, and include changes in drug metabolism, oxidative stress, cytokine profiles and immune hyperactivation.

Nevertheless, mast cell disorders might be ruled out in cases of severe systemic reactions. Careful examination of the skin should accompany measurement of basal serum tryptase levels.
