The effective management of AIDS with HIV protease inhibitors, or the use of angiotensin-converting enzyme inhibitors to treat hypertension, indicates that proteases do make good drug targets. On the other hand, matrix metalloproteinase (MMP) inhibitors from several companies have failed in both cancer and rheumatoid arthritis clinical trials.

AbstractReceptors, located on both the cell surface and within the cell, are the molecular targets through which drugs produce their beneficial effects in various disease states. Receptors were initially conceptualized at the beginning of the 20th century by the parallel efforts of Ehrlich and Langley.

Several classes of clinically important antibiotics target the bacterial ribosome, where they interfere with microbial protein synthesis. Structural studies of the interaction of antibiotics with the ribosome have revealed that these small molecules recognize predominantly the rRNA components.

'Now, if we were to succeed in discovering amongst these (chemoceptors) a receptor which was not represented in the organs of the host, we would have the possibility of constructing an ideal medicament by selecting a haptophore group that fits exclusively this particular receptor of the parasite. A medicament provided with such a haptophore group would

The elucidation of the crystal structure of the ribosome and its subunits has dramatically increased our understanding of this organelle and the molecular interactions that determine its functional capabilities. Two recent publications, one on the structure of the bacterial ribosome at 3.5A resolution and one on the identification of functionally ...

Publisher Summary Antioxidants have been known to prevent lipid peroxidation, the depletion of reduced glutathione and other intracellular protectants against oxidation, and lipoprotein oxidation. Antioxidants help the regulation of signal transduction, kinase and phosphatase activation, gene expression, and enzyme activation.
