Real-time imaging of oxidative and nitrosative stress in the liver of live animals for drug-toxicity testing

open access: yesNature Biotechnology, 2014
Current drug-safety assays for hepatotoxicity rely on biomarkers with low predictive power. The production of radical species, specifically reactive oxygen species (ROS) and reactive nitrogen species (RNS), has been proposed as an early unifying event ...

Soman induces endoplasmic reticulum stress and apoptosis of cerebral organoids via the GRP78‐ATF6‐CHOP signaling pathway

open access: yesFEBS Open Bio, EarlyView.
Cerebral organoids were employed as a novel model to explore the neurotoxicity of soman. Soman inhibited acetylcholinesterase activity, increased cell apoptosis and upregulated endoplasmic reticulum (ER) stress markers glucose‐regulated protein 78 (GRP78), activating transcription factor 6 (ATF6) and C/EBP homologous protein (CHOP).

Multimodal AI predicts clinical outcomes of drug combinations from preclinical data [PDF]

open access: yesarXiv
Predicting clinical outcomes from preclinical data is essential for identifying safe and effective drug combinations. Current models rely on structural or target-based features to identify high-efficacy, low-toxicity drug combinations. However, these approaches fail to incorporate the multimodal data necessary for accurate, clinically-relevant ...

KCS1 and VIP1, the genes encoding yeast phosphoinositol pyrophosphate synthases, are required for Ca2+‐mediated response to dimethylsulfoxide (DMSO)

open access: yesFEBS Open Bio, EarlyView.
Ca2+‐mediated response to DMSO was investigated in Saccharomyces cerevisiae cells expressing Ca2+‐dependent aequorin. Cell exposure to DMSO induced a cytosolic Ca2+ wave dependent on the integrity of the Cch1/Mid1 channel. Deletion of KCS1 or VIP1 genes encoding the phosphoinositol pyrophosphate (PP‐IP) synthases suppressed the DMSO‐induced Ca2 ...

Equivalence tests for binary efficacy-toxicity responses [PDF]

open access: yesarXiv, 2019
Clinical trials often aim to compare a new drug with a reference treatment in terms of efficacy and/or toxicity depending on covariates such as, for example, the dose level of the drug. Equivalence of these treatments can be claimed if the difference in average outcome is below a certain threshold over the covariate range.

Natural calcium carbonate for biomedical applications [PDF]

open access: yesarXiv, 2016
Sea shells are found to be a very rich natural resource for calcium carbonate. Sea shells are made up of CaCO3 mainly in the aragonite form, which are columnar or fibrous or microsphere structured crystals. The bioactivity of nanoparticles of sea shell has been studied in this work. The sea shells collected were thoroughly washed, dried and pulverized.
