Identification and characterisation of two functional antibiotic MATE efflux pumps in the archaeon Halorubrum amylolyticum

open access: yesnpj Antimicrobials and Resistance
Multidrug efflux pumps have been found to play a crucial role in drug resistance in bacteria and eukaryotes. In this study, we investigated the presence of functional multidrug and toxic compound extrusion (MATE) efflux pumps, inferred from whole genome ...

ABCABC Stacking‐Enabled Non‐Centrosymmetry 3R‐ZnIn2S4 Nanosheets for Piezocatalytic Uranium Extraction

open access: yesAdvanced Science, EarlyView.
The ABCABC stacking sequence in 2D ZnIn2S4 nanosheets creates intrinsic non‐centrosymmetry, enabling robust piezocatalysis under weak mechanical agitation. The nanosheets achieve a H2O2 yield of 276.7 µmol g−1 h−1 and uranium extraction capacity of 763.2 mg g−1 via a dual mechanism of direct piezoreduction and in situ H2O2 complexation.

A Programmable Calcification Nanoplatform for Loco‐Regional Calcification‐Immune Hepatocellular Carcinoma Therapy

open access: yesAdvanced Science, EarlyView.
An in situ‐grown BP‐CaO2 nanoplatform supplies coordinated Ca2+, endogenous phosphate, and oxidative stress to convert tumor calcification from a passive endpoint into an active immune‐remodeling process. Widespread hydroxyapatite deposition is visualized by CT, while multi‐omics reveals MCOLN2 as a calcium‐responsive mediator linking biomineralization

Reversing Temozolomide Resistance in Glioblastoma Based on Cuproptosis‐Mediated Positive Feedback Loop

open access: yesAdvanced Science, EarlyView.
To effectively reverse TMZ resistance, this work proposes a cuproptosis‐mediated positive feedback loop that integrates mitochondrial dysfunction, ATP depletion, AMPK‐p53 signaling, and glycolytic inhibition, ultimately amplifying cell death. This cuproptosis‐mediated feedback loop simultaneously disrupts multiple resistance pathways in GBM, including ...

Pyrazole‐Derived Antibacterial Compounds Effectively Treat Methicillin‐Resistant Staphylococcus Aureus Infections by Inhibiting Aspartate Transcarbamoylase

open access: yesAdvanced Science, EarlyView.
This study unveils a novel antibacterial mechanism against MRSA, in which the compound Py‐27 selectively targets and inhibits aspartate transcarbamoylase (ATCase), a key enzyme in pyrimidine synthesis. This inhibition disrupts DNA replication, induces oxidative damage, leading to potent bactericidal activity.

Efflux Pumps Role in the Emergence of MDR Bacteria [PDF]

open access: yes
The emergence multidrug tolerance is the most threatening obstacle for the population health.  During the last years, transporting pumps are implicated in the emergence of antibiotic resistance.

Asymmetric Single‐Atom Catalysts Govern Diffusible and Non‐Diffusible Non‐Radical Pathways for Selective Degradation of Antibiotic Resistance Genes

open access: yesAdvanced Science, EarlyView.
Asymmetric single‐atom metal‐nitrogen‐carbon catalysts modulate the non‐radical pathway during peroxymonosulfate (PMS) activation. A fundamental contrast was revealed between diffusible singlet oxygen (1O2) and non‐diffusible electron‐transfer pathways (ETP) in antibiotic resistance gene (ARG) degradation.

Understanding the role of multidrug efflux pumps in Shigella

open access: yes, 2018
The efflux pumps represent an important and consistent group of proteins involved in transport process present in all living cells. The importance of efflux pumps in bacteria has always been associated to their ability to extrude a wide range of ...
