Circulating elastin peptides, role in vascular pathology

The atherosclerotic process starts with the degradation of elastic fibers. Their presence was demonstrated in the circulation as well as several of their biological properties elucidated. We described years ago a procedure to obtain large elastin peptides by organo-alkaline hydrolysis, κ-elastin.

Abstract Two fractions have been isolated from extracts obtained by the autoclave treatment of purified elastin from bovine ligaments. One fraction contains a very low proportion of polar amino acids and no cross-links. The second fraction is relatively rich in polar amino acids and in lysinonorleucine.

Effect of Elastin Peptides on Vascular Tone

As elastin receptors exist on several cell types, especially endothelial cells, this investigation was carried out to study the effect of elastin peptides on vascular tone. For this purpose, rat aortic rings were mounted in an organ bath for isometric tension measurements.

Guanidination of the Lysine Residues Present in Fibrous Elastin and in Soluble Elastin Peptides

We have found that only 20 to 30% of the lysine residues measured after acid hydrolysis of insoluble elastin and alpha-elastin were blocked by prior guanidination of elastin with o-methylisourea. In contrast free lysine was guanidinated almost 100% and a purified, highly crosslinked peptide fraction of low molecular weight, 85%.

ANTIGENICITY OF HUMAN LUNG ELASTIN PEPTIDES

Antigenic determinants of human lung elastin peptides were studied by fractionation on an SE-Sephadex C-50 column. By elution profile, tubes were pooled into 12 fractions. Amino acid analysis of these fractions showed that the first seven contained mostly non-polar peptides which lacked desmosine and isodesmosine residues.
