Capture and Culture of Circulating Tumor Cells Using a Biodegradable 3D Scaffold for Patient‐Derived Xenograft Construction

open access: yesAdvanced Science, EarlyView.
A self‐supporting, biocompatible, and biodegradable 3D cellulose scaffold has been developed for efficient isolation and in situ expansion of circulating tumor cells (CTCs) from patient blood. Subsequently, the 3D cellulose scaffold with cultured CTCs is directly implanted into immunodeficient mice for constructing patient CTC‐derived xenograft (CDX ...

B4GALNT4‐Mediated Glycosylation of PDK1 Activates the PI3K‐AKT Signaling Pathway to Promote Prostate Cancer Progression

open access: yesAdvanced Science, EarlyView.
This study identifies B4GALNT4 as a key driver of prostate cancer (PCa). It shows that B4GALNT4 glycosylates PDK1 protein at the N531 site, which stabilizes the PDK1 and constitutively activates the PI3K‐AKT pathway. This mechanism promotes tumor cell proliferation, migration, and invasion. The findings establish the B4GALNT4‐PDK1 glycosylation axis as

EMT and Cancer: More Than Meets the Eye.

open access: yesDevelopmental Cell, 2019
Epithelial cells acquire mesenchymal characteristics during development, wound healing and inflammation, and in cancer and fibrosis. With increasing appreciation of different roles of epithelial-mesenchymal transition (EMT), we address the question of ...

PDZK1‐ULK1 Axis Triggers Lipophagy to Inhibit Tumor Progression and Sunitinib Resistance in Clear Cell Renal Cell Carcinoma

open access: yesAdvanced Science, EarlyView.
This study reveals that PDZK1 loss impairs ULK1‐mediated lipophagy, driving lipid droplet accumulation and sunitinib resistance in clear cell renal cell carcinoma. PDZK1 inhibits LEF1 nuclear translocation, enhancing ULK1 transcription. Pharmacological ULK1 activation resensitizes resistant tumors to sunitinib.

Quantifying the photothermal conversion efficiency of plasmonic nanoparticles by means of terahertz radiation

open access: yesAPL Photonics, 2019
The accurate determination of the photothermal response of nanomaterials represents an essential aspect in many fields, such as nanomedicine. Specifically, photothermal cancer therapies rely on the precise knowledge of the light-to-heat transfer ...

EMT is not required for lung metastasis but contributes to chemoresistance

open access: yesNature, 2015
The role of epithelial-to-mesenchymal transition (EMT) in metastasis is a longstanding source of debate, largely owing to an inability to monitor transient and reversible EMT phenotypes in vivo.

NIR‐Activatable Domino Cascade Catalysis Nanozyme Reactor for Multi‐Mechanism Synergistic Immunotherapy in Bladder Cancer

open access: yesAdvanced Science, EarlyView.
This study presents an intravesical delivery strategy of ferroptosis‐ and immunity‐inducing nanozyme‐loaded thermoresponsive hydrogels (FMCC@PNH) for enhanced bladder cancer therapy. Abstract Current intravesical therapies for bladder cancer after resection are limited by poor tissue penetration, off‐target effects, and insufficient efficacy.

Allogeneic Immune Cell Perfusion Inhibits the Growth of Vascularized 3D In Vitro Tumor Models, Induces Vascular Regression and Desmoplasia, but Promotes Tumor Cell Invasion

open access: yesAdvanced Science, EarlyView.
This study presents a vascularized 3D tumor model to investigate immune–stromal–tumor interactions under allogeneic PBMC perfusion. While immune cells induced tumor shrinkage, they also promoted vascular regression, stromal activation, and cancer cell invasion.

DSG2+ Cancer Stem Cells Co‐Located With FAP+ Myofibroblasts in the Tumor Boundary That Determines the Efficacy of Immunotherapy in Non‐Small Cell Lung Cancer

open access: yesAdvanced Science, EarlyView.
Cancer stem cells (CSCs) in non‐small cell lung cancer display pronounced plasticity and spatial heterogeneity. By integrating single‐cell and spatial transcriptomics, this study defines a DSG2‐associated CSC program and reveals a tumor‐margin niche formed with FAP+ myofibroblasts.
