Lipid Nanoparticle‐Mediated CRISPR‐Cas13a Delivery for the Control of Bacterial Infection

open access: yesAdvanced Healthcare Materials, Volume 14, Issue 7, March 14, 2025.
New formulations of lipid nanoparticles (LNPs) that can deliver nucleic acids to Gram‐negative bacteria are proposed to combat bacterial infection. The delivery of nucleic acids by LNPs is aided by LNP‐helpers which weaken the bacterial outer membrane. LNPs encapsulating the Cas13a/gRNA expression vector achieve an antibacterial effect in both in vivo ...

Pseudomonas aeruginosa gene PA4880 encodes a Dps-like protein with a Dps fold, bacterioferritin-type ferroxidase centers, and endonuclease activity

open access: yesFrontiers in Molecular Biosciences
We report the biochemical, structural, and functional characterization of the protein coded by gene PA4880 in the P. aeruginosa PAO1 genome. The PA4880 gene had been annotated as coding a probable bacterioferritin.

Characterization of endonuclease activities in Moloney murine leukemia virus and its replication-defective mutants [PDF]

open access: yes, 1987
To study Moloney murine leukemia virus (M-MulV) proteins associated with the integration of proviral DNA into the host chromosome, we isolated endonuclease activities from purified virion preparations of the wild type and two of its replication mutants ...

Endonuclease II of coliphage T4: a recombinase disguised as a restriction endonuclease? [PDF]

open access: yesMolecular Microbiology, 1998
EndoII shares with restriction endonucleases the property of cleaving foreign DNA while leaving the endonuclease‐encoding genome intact, ensuring the survival of one DNA species in the cell. In addition, in vivo EndoII cleaves a specific DNA sequence and cleavage is context dependent. These context effects extend over at least 1000 bp, largely limiting

The Diversity of Ribonuclease P: Protein and RNA Catalysts with Analogous Biological Functions

open access: yesBiomolecules, 2016
Ribonuclease P (RNase P) is an essential endonuclease responsible for catalyzing 5’ end maturation in precursor transfer RNAs. Since its discovery in the 1970s, RNase P enzymes have been identified and studied throughout the three domains of life ...

Rapid identification and differentiation of the vaccine strain Rac H from EHV 1 field isolates using a non-radioactive DNA probe [PDF]

open access: yes, 1992
A method for rapid differentiation between the EHV 1 live vaccine strain Rac H and field isolates is described. Total DNA was isolated from virus-infected small scale cell cultures.

Dissecting endonuclease and exonuclease activities in endonuclease V from Thermotoga maritima [PDF]

open access: yesNucleic Acids Research, 2010
Endonuclease V is an enzyme that initiates a conserved DNA repair pathway by making an endonucleolytic incision at the 3'-side 1 nt from a deaminated base lesion. DNA cleavage analysis using mutants defective in DNA binding and Mn(2+) as a metal cofactor reveals a novel 3'-exonuclease activity in endonuclease V [Feng,H., Dong,L., Klutz,A.M ...

Engineering CRISPR System‐Based Bacterial Outer Membrane Vesicle Potentiates T Cell Immunity for Enhanced Cancer Immunotherapy

open access: yesAdvanced Materials, EarlyView.
This study develops a multifunctional CRISPR‐dCas9‐based OMV platform termed OMV‐C9I12, which facilitates the coexpression of CXCL9 and IL‐12 within tumor cells. This platform enhances T cell recruitment and activation, synergizes with anti‐PD‐1/PD‐L1 immunotherapy, and amplifies antitumor T cell immunity.
