Effects of oxidative stress on the solubility of HRD1, a ubiquitin ligase implicated in Alzheimer's disease.

open access: yesPLoS ONE, 2014
The E3 ubiquitin ligase HRD1 is found in the endoplasmic reticulum membrane of brain neurons and is involved in endoplasmic reticulum-associated degradation. We previously demonstrated that suppression of HRD1 expression in neurons causes accumulation of

Metastasis on pause: How dormant tumor cells stay hidden within the tumor microenvironment and evade immune surveillance

open access: yesMolecular Oncology, EarlyView.
Dormant cancer cells can hide in distant organs for years, evading treatment and the immune system. This review highlights how signals from the surrounding tissue and immune environment keep these cells inactive or trigger their reawakening. Understanding these mechanisms may help develop therapies to eliminate or control dormant cells and prevent ...

Penfluridol induces endoplasmic reticulum stress leading to autophagy in pancreatic cancer

open access: yesTumor Biology, 2017
Pancreatic cancer is one of the most aggressive and difficult to treat cancers. Experimental and clinical evidence suggests that high basal state autophagy in pancreatic tumors could induce resistance to chemotherapy.

A Whole-Genome RNA Interference Screen Reveals a Role for Spry2 in Insulin Transcription and the Unfolded Protein Response. [PDF]

open access: yes, 2017
Insulin production by the pancreatic β-cell is required for normal glucose homeostasis. While key transcription factors that bind to the insulin promoter are known, relatively little is known about the upstream regulators of insulin transcription.

Targeting TNBC: core–shell polycationic polyurea dendrimers with inherent anticancer activity

open access: yesFEBS Open Bio, EarlyView.
Core–shell polycationic PURE dendrimers were tested in TNBC‐derived tumor models. Both formulations selectively targeted TNBC and effectively reduced tumor volume. PUREG4‐OEI48 suppressed tumor growth without detectable toxicity, whereas PUREG4‐OCEI24, despite showing efficacy, induced hepatic toxicity.

Endoplasmic reticulum stress and eIF2α phosphorylation: The Achilles heel of pancreatic β cells

open access: yesMolecular Metabolism, 2017
Background: Pancreatic β cell dysfunction and death are central in the pathogenesis of most if not all forms of diabetes. Understanding the molecular mechanisms underlying β cell failure is important to develop β cell protective approaches.

EXPRESSION OF ENDOPLASMIC RETICULUM OXIDOREDUCTASES (EROS) AND THEIR ROLE IN THE GI TRACT [PDF]

open access: yes, 2012
It has been shown that some ER redox enzymes are differentially expressed in stomach and oesophagus tissue. The tissues of the gastrointestinal system, which are subject to external changes of environment during the process of digestion, represent a ...

The proteasome biogenesis regulator Rpn4 cooperates with the unfolded protein response to promote ER stress resistance

open access: yes, 2019
Misfolded proteins in the endoplasmic reticulum (ER) activate the unfolded protein response (U PR), which enhances protein folding to restore homeostasis.

Micro(RNA)managing Endoplasmic Reticulum Stress [PDF]

open access: yesIUBMB Life, 2013
AbstractCellular disturbances that cause accumulation of misfolded proteins in the endoplasmic reticulum (ER) lead to a condition referred to as “ER stress” and trigger the unfolded protein response (UPR), a signaling pathway that attempts to restore ER homeostasis.

Microglial dynamics and ferroptosis induction in human iPSC‐derived neuron–astrocyte–microglia tri‐cultures

open access: yesFEBS Open Bio, EarlyView.
A tri‐culture of iPSC‐derived neurons, astrocytes, and microglia treated with ferroptosis inducers as an Induced ferroptosis model was characterized by scRNA‐seq, cell survival, and cytokine release assays. This analysis revealed diverse microglial transcriptomic changes, indicating that the system captures key aspects of the complex cellular ...
