Dysfunction of Endothelial Nitric Oxide Synthase and Atherosclerosis [PDF]

open access: yesArteriosclerosis, Thrombosis, and Vascular Biology, 2004
Atherosclerosis is associated with an impairment of endothelium-dependent relaxations, which represents the reduced bioavailability of nitric oxide (NO) produced from endothelial NO synthase (eNOS). Among various mechanisms implicated in the impaired EDR in atherosclerosis, superoxide generated from dysfunctional eNOS has attracted attention.

Novel features of nitric oxide, endothelial nitric oxide synthase, and atherosclerosis

Current Atherosclerosis Reports, 2004
There is a complex pathophysiologic scenario involving nitric oxide (NO), endothelial nitric oxide synthase (eNOS), and the development of atherosclerosis and unstable atheroma. Endothelial damage induced by atherosclerosis leads to the reduction in bioactivity of eNOS with subsequent impaired release of NO.

Endothelial nitric oxide synthase and endothelial dysfunction

Current Hypertension Reports, 2003
Nitric oxide (NO) regulates vascular tone and local blood flow, platelet aggregation and adhesion, and leukocyte-endothelial cell interactions. Abnormalities in NO production by the vascular endothelium result in endothelial dysfunction, which occurs in hypertension, diabetes, aging, and as a prelude to atherosclerosis.

Modulation of endothelial nitric oxide synthase by fibronectin

Molecular and Cellular Biochemistry, 2008
Nitric oxide (NO) produced by the action of endothelial nitric oxide synthase (eNOS) plays an important role in the regulation of vascular tone, cell survival, and angiogenesis. Interaction of endothelial cells (ECs) with a fibronectin (FN) rich matrix is important in the regulation of EC function and survival during angiogenesis. The present study was

Expression and Regulation of Endothelial Nitric Oxide Synthase

Trends in Cardiovascular Medicine, 1997
Endothelium-derived nitric oxide (NO) is a key determinant of blood pressure homeostasis and platelet aggregation and is synthesized by the endothelial isoform of nitric oxide synthase (eNOS). In the vascular wall, eNOS is activated by diverse cell-surface receptors and by increases in blood flow, and the consequent generation of NO leads to vascular ...

Endothelial Nitric Oxide Synthase Polymorphism in Preeclampsia

Journal of the Society for Gynecologic Investigation, 2003
We wished to determine whether genetic variability in the gene encoding endothelial nitric oxide synthase (eNOS) modifies individual susceptibility to the development of preeclampsia. The study involved 132 preeclamptic and 113 healthy control pregnant women who were genotyped for the Glu298Asp polymorphism in the eNOS gene.

Regulation of Endothelial Nitric Oxide Synthase in Cardiac Remodeling

SSRN Electronic Journal, 2021
Our previous study demonstrated that endothelial nitric oxide synthase (eNOS) gene serves as a candidate for modifiers of hypertrophic cardiomyopathy (HCM), which alters severity of HCM phenotypes. Herein, we sought to further elucidate the role of eNOS on cardiac myocyte hypertrophy and fibrosis, the major phenotypes of HCM.Male eNOS-deficient mice ...

Nitric Oxide in Obstructive Uropathy: Role of Endothelial Nitric Oxide Synthase

The Journal of Urology, 2002
Nitric oxide (NO) ameliorates fibrosis in experimental obstructive uropathy. Previously, we demonstrated that renal fibrosis was decreased after 2 weeks of unilateral ureteral obstruction in inducible nitric oxide synthase (iNOS) knock-out mice given L-arginine supplemented drinking water.

Direct and reversible inhibition of endothelial nitric oxide synthase by nitric oxide

American Journal of Physiology-Heart and Circulatory Physiology, 1995
The objective of this study was to investigate the regulation of endothelial nitric oxide (NO) synthase by NO. Partially purified endothelial NO synthase was exposed to authentic NO (10-200 microM) and to the nitrovasodilators sodium nitroprusside (SNP; 10-1,000 microM) and S-nitroso-N-acetylpenicillamine (SNAP; 100-1,000 microM), and enzyme activity ...

Cellular regulation of endothelial nitric oxide synthase

American Journal of Physiology-Renal Physiology, 2001
Renal function is highly dependent on endothelium-derived nitric oxide (NO). Several renal disorders have been linked to impaired NO bioavailability. The enzyme that is responsible for the synthesis of NO within the renal endothelium is endothelial NO synthase (eNOS). eNOS-mediated NO generation is a highly regulated cellular event, which is induced by
