Soluble SORL1 in cerebrospinal fluid as a marker for functional impact of rare SORL1 variants

open access: yesAlzheimer's &Dementia, Volume 22, Issue 2, February 2026.
Abstract INTRODUCTION The sortilin‐related receptor (SORL1) directs APP and Aβ trafficking within the retromer pathway. Cleavage at the cell surface releases soluble SORL1 (sSORL1) into cerebrospinal fluid (CSF). We examined whether CSF‐sSORL1 can serve as an in vivo marker of genetically impaired SORL1.

Factors impacting survival in individuals with Down syndrome‐associated Alzheimer's disease

open access: yesAlzheimer's &Dementia, Volume 22, Issue 2, February 2026.
Abstract INTRODUCTION Adults with Down syndrome (DS) are at high risk for Alzheimer's disease (AD), the leading cause of death in this population. Survival in DS after AD diagnosis appears shorter than in sporadic AD; however, the factors influencing survival remain poorly understood.

Temporal dynamics of white matter hyperintensities related to Alzheimer's disease in adults with Down syndrome

open access: yesAlzheimer's &Dementia, Volume 22, Issue 2, February 2026.
Abstract INTRODUCTION White matter hyperintensities (WMH) are common in Down syndrome (DS), yet their longitudinal evolution and associations with Alzheimer's disease (AD) remain unclear. METHODS Longitudinal MRI study, including 80 DS adults and 53 euploid controls. WMH were segmented on serial FLAIR using a longitudinal pipeline.

Cognitive profiles at presentation and subsequent cognitive decline and quality of life in Parkinson's disease

open access: yesAlzheimer's &Dementia, Volume 21, Issue 12, December 2025.
Abstract INTRODUCTION Currently, our ability to predict cognitive decline in Parkinson's disease is limited. METHODS In 234 dementia‐free PD patients, cognitive profiles at baseline were determined using cluster analysis and longitudinal associations with global cognitive z‐score, health‐related quality of life (HR‐QoL), caregiver burden, and risk of ...

Association of BMI and Cognitive Performance in the Diabetes Prevention Program Outcomes Study

open access: yesObesity, Volume 33, Issue 12, Page 2365-2376, December 2025.
ABSTRACT Objective This study aimed to examine the association of BMI with cognitive performance in individuals with diabetes or prediabetes. Methods Among Diabetes Prevention Program Outcomes Study (DPPOS) participants, BMI was categorized as normal (< 25 kg/m2), overweight (25 to < 30 kg/m2), or obesity (≥ 30 kg/m2).

Cortical thickness subtypes in cognitively unimpaired individuals: Differential network and transcriptomic vulnerability to cortical thinning

open access: yesAlzheimer's &Dementia, Volume 21, Issue 10, October 2025.
Abstract INTRODUCTION The emergence, stability, and contributing factors of Alzheimer's disease (AD) gray matter subtypes remain unclear. METHODS We analyzed data from 1323 individuals without a diagnosis of dementia (CDR < 1) with T1w‐MRI and amyloid‐PET, including 622 with longitudinal data (3.66 ± 1.78 years).
