Embryo‐like structures (stembryos) are an innovative tool, but they are hindered by experimental variability and limited developmental potential. DNA methylation is crucial for mammalian development, but its status in stembryo models is poorly characterized.

pH‐mediated activation of the lysosomal arginine sensor SLC38A9
Cells monitor nutrient levels via the lysosomal transporter SLC38A9 to activate the mechanistic target of rapamycin complex 1 (mTORC1). This study reveals that SLC38A9 function is regulated by pH. We identified histidine 544 as a critical pH sensor that undergoes conformational changes to control amino acid efflux from lysosomes; therefore, it ...

Enzyme Inhibition in Relation to Chemotherapy.
Summary1. The effect of enzyme concentration on the inhibition produced by certain inhibitors of the succinic dehydrogenase system has been experimentally determined. The inhibitors studied were malonate, itaconate, oxalacetate, quinone and cupric ion.2. The reaction between succinic dehydrogenase and copper or quinone was not immediate but required 30

Degradation mechanism of the von Willebrand factor A2 domain by nattokinase
Nattokinase, a natto‐derived protease, exhibits potent antithrombotic effects. This study demonstrates that nattokinase directly cleaves the von Willebrand factor (vWF) A2 domain in vitro. Unlike the native regulator ADAMTS13, nattokinase degrades folded vWF independently of shear stress.

Inhibition of enzymes by alkylsalicylic acids
Abstract5-n-Alkylsalicylates inhibited a variety of enzymes that transform acidic substances, viz. glucose-6-phosphate dehydrogenase, glyoxalase, xanthine oxidase, carbonic anhydrase and D-amino-acid oxidase. Inhibitory potency rose to a peak at the n-nonyl derivative.

The ubiquitin‐proteasome system and autophagy as guardians of the cellular proteome
This Perspective covers the three principles governing the crosstalk between the ubiquitin‐proteasome system and autophagy in cellular proteostasis: (1) a shared ubiquitin code routing substrates via shuttle factors or autophagy receptors; (2) spatial compartmentalization into phase‐separated degradation hubs and organelle‐specific modules (exemplified

An unexpected alternative interaction site for ethyl viologen was identified in formate dehydrogenase 1 from Methylorubrum extorquens. Combined mutagenesis, kinetic analysis, and docking revealed that aromatic residues near an iron–sulfur cluster enable flavin mononucleotide‐independent electron transfer, offering a framework for engineering improved ...
