Engineering enzymes for stability
Current Opinion in Structural Biology, 1996There have been many recent developments in elaborating the approaches for stabilizing enzymes by stabilizing the folding state, destabilizing the unfolded state and altering the kinetics of unfolding. However, these represent a series of rules of thumb rather than the reliable principles that would be expected of 'engineering'.

The enzyme stability of dehydropeptides
Biochemical and Biophysical Research Communications, 1978Abstract A pentapeptide, Z-Gly-Gly-Phe-Phe-Ala · OH (1b) and the corresponding unsaturated pentapeptide, Z-Gly-Gly-Phe-Δ Z Phe-Ala · OH (1a) , have been synthesized. The saturated compound (1b) was rapidly hydrolyzed by both chymotrypsin and thermolysin to the expected products, but the dehydropeptide was completely unhydrolyzed by ...

Enzyme stability in downstream processing part 1: Enzyme inactivation, stability and stabilization
Biotechnology Advances, 1992In biotechnological recovery processes the instability of the product can lead to large losses in the sequence of recovery processes needed to purify the product. As the cost of the final active product is strongly dependent on the recovery yield, this will lead to an increase in product cost.

Stabilization of enzymes by their specific antibodies
Enzyme and Microbial Technology, 1991In nature, increased stability of enzymes has often been found to be associated with noncovalent protein-protein interactions. Specific antibodies should be suitable for this purpose. To test this hypothesis, we used a number of model enzymes, complexed them with their specific antibodies, and exposed them and the free enzymes to low and high ...

Rational engineering of enzyme stability
Journal of Biotechnology, 2004During the past 15 years there has been a continuous flow of reports describing proteins stabilized by the introduction of mutations. These reports span a period from pioneering rational design work on small enzymes such as T4 lysozyme and barnase to protein design, and directed evolution.

Structural stability of an enzyme biocatalyst
Biochemical Society Transactions, 2007TK (transketolase) undergoes inactivation during biocatalytic processes due to oxidation, substrate and product inhibition, reactivity of aldehyde substrates, irreversible inactivation at low pH, and dissociation of cofactors. However, the contribution of protein denaturation to each of these mechanisms is not fully understood.

Stabilization of Enzymes by Using Thermophiles
2023Manufactured steroid compounds have many applications in the pharmaceutical industry. Due to the chemical complexity and chirality of steroids, there is an increasing demand for enzyme-based bioconversion processes to replace those based on chemical synthesis.

The enzyme stability of dehydro-enkephalins
Peptides, 1982Dehydro-enkephalins [delta Ala2]-, [delta Ala3]-, [delta Phe4]-, and [delta Leu5]enkephalins, were examined for their stability to enzymatic hydrolysis by carboxypeptidase Y [EC 3.4.16.1]. The successively liberated amino acids were determined quantitatively by amino acid analyses.

Effect of Salts on Enzyme Stability
