Prostanoid EP₂ Receptors Are Up-Regulated in Human Pulmonary Arterial Hypertension: A Key Anti-Proliferative Target for Treprostinil in Smooth Muscle Cells [PDF]

open access: yes, 2018
Prostacyclins are extensively used to treat pulmonary arterial hypertension (PAH), a life-threatening disease involving the progressive thickening of small pulmonary arteries. Although these agents are considered to act therapeutically via the prostanoid

Biomimetic Macrophage Cell Membrane‐Based Nanoparticles for Effective Treatment of Glioblastoma Through Boron Neutron Capture Therapy Combined With Immunotherapy

open access: yesAdvanced Science, EarlyView.
ABSTRACT Glioblastoma multiforme (GBM) remains largely incurable due to the blood‐brain barrier (BBB) and immunosuppressive microenvironment. While boron neutron capture therapy (BNCT) selectively eradicates tumor cells via 10B(n, α)‐7Li reactions, its clinical potential in GBM is unrealized because of the suboptimal pharmacokinetics of conventional ...

Prostaglandin E𝟐 and the Suppression of Phagocyte Innate Immune Responses in Different Organs

open access: yesMediators of Inflammation, 2012
The local and systemic production of prostaglandin E2 (PGE2) and its actions in phagocytes lead to immunosuppressive conditions. PGE2 is produced at high levels during inflammation, and its suppressive effects are caused by the ligation of the E ...

Pathophysiological role of extrasynaptic GABAA receptors in typical absence epilepsy [PDF]

open access: yes, 2011
GABA is the principal inhibitory neurotransmitter in the mammalian CNS. It acts via two classes of receptors, the GABAA, a ligand gated ion channel (ionotropic receptor) and the metabotropic G-protein coupled GABAB receptor.

CD177 Deficiency Defines a Stable Subtype of Human Neutrophil Granulocytes with Tumor Promoting Activity

open access: yesAdvanced Science, EarlyView.
Human neutrophils exist as two epigenetically imprinted subtypes defined by stable CD177 expression or absence — a ratio that persists across time, circadian rhythms, and inflammation. CD177− neutrophils display a distinct molecular landscape enriched in arginase 1 and lipid metabolism markers, accumulate in head‐and‐neck tumors, and associate with ...

Ultra‐Radiostable Covalent Conformationally Interlocked Networks Enabling a Universal Radiometal‐Labeling Platform for Cancer Radioembolization

open access: yesAdvanced Science, EarlyView.
Conjugated poly(imide dioxime)‐based microspheres establish a radiometal coordination‐driven conformational interlocked network with ultra‐high radiostability. This platform enables low‐temperature, multi‐radionuclide labeling for SPECT/PET/MRI imaging and radionuclide therapy. Mechanistic insights from EXAFS and DFT reveal enhanced stability, while in

THBS1+ Macrophages Exacerbate Modic Changes via SDC4‐Dependent Activation of NLRP3 Inflammasome

open access: yesAdvanced Science, EarlyView.
The illustration of THBS1+ macrophages exacerbate MCs via SDC4‐dependent activation of the NLRP3 inflammasome. THBS1+ macrophage subpopulation was identified in MCs through single‐cell RNA sequencing. C. acnes and its metabolites activate THBS1 expression in macrophages via the TLR signaling pathway.

Molecular and Cellular Hallmarks of Age‐Related Vestibular Hair Cell Degeneration

open access: yesAdvanced Science, EarlyView.
This study utilizes single‐cell RNA‐seq transcriptomes, advanced imaging, and electrophysiology to examine universal and cell‐type‐specific aging signatures of vestibular hair cells. The study shows that impaired hair bundle function is a key driver of age‐related vestibular dysfunction.
