Genetic encoding of noncanonical amino acids (ncAA) enables their site‐specific installation in proteins. This work compares the nuclear magnetic resonance (NMR) performance of different ncAAs in a large protein−DNA complex (~ 100 kDa) without isotope labelling. The ncAAs deliver readily detectable NMR signals in the complex between E.

Evolutionary and mechanistic divergence in the Hsp70–Hsp110 chaperone system. Prokaryotic Hsp70s probably diversified into multiple orthologues that cooperated with co‐chaperones such as JDPs and NEF, forming increasingly complex proteostasis networks.

Recombinant Fibrinogen-Gamma-Chain as a Crosslinker of Thiol-ene Hydrogels. [PDF]
Fibrinogen gamma chain (FGG) was recombinantly expressed in E. coli to generate a recombinant protein containing free thiol groups from cysteine. The expressed protein was used to crosslink norbornene functionalized gelatin providing a proof‐of‐principle of this novel crosslinker type and the formed hydrogels were characterized for their stiffness ...

The genes PH0137, PH0138, and PH0140 form a cluster in the genome of Pyrococcus horikoshii. The PH0140 gene encodes a transcription factor identified as a d‐amino acid‐responsive regulatory protein (DARP). In feast mode, l‐Ile‐bound DARP binds to the promoter and represses transcription of PH0137 and PH0138. In famine mode, DARP binds to d‐allo‐Ile and

DNA aptamers, Apt 60 and Apt 173, target the BamA protein to compromise outer membrane integrity in Pseudomonas aeruginosa, making it susceptible to Azithromycin, an antibiotic that is otherwise poorly effective due to the low‐permeability membrane.

Fragile X messenger ribonucleoprotein 1 (FMRP) is a multidomain RNA‐binding protein associated with Fragile X Syndrome (FXS). We found that its N‐terminal structured region has an intrinsic propensity to undergo liquid–liquid phase separation and fibril formation. FXS‐associated mutations perturb protein stability and aggregation propensity, suggesting

Cyclic β‐1,2‐glucan (CβG)‐associated solute‐binding protein of type I ABC transporter was originally identified in Chloroflexus aurantiacus, a phototrophic bacterium. Crystal structures in complex with CβGs revealed unique binding modes that are fundamentally distinct from that of the short β‐1,2‐glucooligosaccharides‐binding protein.

Hulless barley resists the newly identified sheath rot disease caused by the necrotrophic fungal pathogen Dactylobotrys graminicola by releasing coniferyl aldehyde, a natural compound that disrupts energy production in the fungus. ABSTRACT Dactylobotrys graminicola (Dgr), a necrotrophic fungal pathogen, has recently been identified as the causative ...

The endoplasmic reticulum (ER) is a key structure in plant cells that helps produce and transport proteins. We discovered that different Lunapark proteins shape ER structures in distinct ways, affecting protein movement and secretion. Understanding how ER structure influences function can help improve plant‐based production of valuable compounds, such ...
