A Dynamic Gate Enables Regioselective Hydroxylation of Free Arginine by a Non‐Canonical Heme Enzyme
The YqcI/YcgG enzyme AglA employs a unique “sandwich” fold and a dynamic gating mechanism to selectively hydroxylate AMPn‐Arg at Nε. Site‐directed mutations reprogram regioselectivity or enable activity on free arginine by altering hydrogen‐bond networks, precisely positioning substrates.

This study revealed that low expression of YTHDC1 in TMZ‐resistant GBM cells leads to increased FSCN1 expression, which is suppressed by m6A modification. FSCN1 activates the CDC42/N‐WASP/Arp2/3 axis in the cell nucleus by recruiting the guanine nucleotide exchange factor (GEF) family member FGD1, thereby promoting F‐actin polymerization in the nucleus.

This study identifies TOPK as a dual‐function target in gastric cancer: it drives tumor cell proliferation and migration, and under IFN‐γ stimulation, phosphorylates eIF4F complex component eIF4A1 to enhance STAT1 mRNA translation, thereby promoting PD‐L1/IDO1‐mediated tumor immunometabolic evasion.
