CircZNF148 stabilizes HK1 through deubiquitinase recruitment, thereby enhancing glycolysis and lactate production. Elevated lactate promotes PD‑L1 lactylation and membrane accumulation while suppressing CD8+ T‐cell cytotoxicity, collectively facilitating immune evasion and malignant progression in TNBC.

The human Werner syndrome protein (hWRN-p) possessing DNA helicase and exonuclease activities is essential for genome stability. Plants have no homologue of this bifunctional protein, but surprisingly the Arabidopsis genome contains a small open ...

A liquid‐phase ion trap (LPIT) coupled with ESI‐MS enables rapid, high‐fidelity de novo sequencing of long oligonucleotides in ∼30 min. By applying an electric field to dissociate metal adducts in solution, LPIT delivers clean spectra, superior mass accuracy, and full sequence coverage, surpassing MALDI‐TOF and LC‐MS.

Molecular snapshots of APE1 proofreading mismatches and removing DNA damage
The essential DNA repair enzyme apurinic/apyrimidinic endonuclease 1 (APE1) has both endonuclease and exonuclease activities. Here, the authors present DNA bound human APE1 crystal structures which give insights into its exonuclease mechanism.

An m6A‐modified circRNA, circPSMB1, is identified as a tumor suppressor in chordoma. METTL3‐dependent m6A promotes circPSMB1 interaction with HNRNPA2B1, limiting c‐MYC expression and malignant progression. An injectable HMnO2‐based nanohydrogel enables local circPSMB1 delivery and tumor microenvironment‐responsive therapy, offering a promising strategy

This study characterizes MfAgo, an enzyme exhibiting highly efficient DNA‐guided DNA and RNA cleavage capability. Precise RNA cleavage strictly requires dimerization to stabilize the PIWI‐loop region, while its robust DNA cleavage functions in a dimerization‐independent manner.

Structural and functional investigation of DinG containing a 3′–5′ exonuclease domain
Damage-inducible gene G (DinG), a bacterial homolog of SF2 helicase, has been extensively studied in Escherichia coli. However, the structural and functional characteristics of DinG homologs fused with an N-terminal 3′–5′ exonuclease domain, such as ...

Exonuclease-enhanced prime editors [PDF]
Prime editing (PE) is a powerful gene-editing technique based on targeted gRNA-templated reverse transcription and integration of the de novo synthesized single-stranded DNA. To circumvent one of the main bottlenecks of the method, the competition of the

Human DNA polymerase delta requires an iron–sulfur cluster for high-fidelity DNA synthesis
The iron–sulfur cluster in human DNA polymerase delta has an impact on DNA polymerase and exonuclease activities and can hence influence the fidelity of DNA synthesis. Replication of eukaryotic genomes relies on the family B DNA polymerases Pol α, Pol δ,
