Structure‐guided design of a sulfamoyl‐modified analog of GS‐441524 yielded a selective, high‐affinity inhibitor of the SARS‐CoV‐2 macrodomain Nsp3b. The compound occupies the phosphate‐binding pocket, forming a stabilizing hydrogen‐bond network that enhances binding affinity (K_D = 0.86 μM).

This study explores structural tuning of SARS‐CoV‐2 Mpro inhibitors through P2 position substitutions and novel warheads. Combining computational modeling and biochemical assays, we found that hydrophobic π‐rich groups and uncommon warheads like fluoro‐vinylsulfone or nitrile enhance potency.

A 4-month-old female entire domestic shorthair cat presented with an acute onset of blindness, tetraparesis and subsequent generalised seizure activity. Haematology and serum biochemistry demonstrated a moderate, poorly regenerative anaemia, hypoalbuminaemia and hyperglobulinaemia with a low albumin:globulin ratio.
