Studying Phosphoroorganic Modification of Smac‐Mimetics to Boost Selectivity for cIAP1‐BIR3 and Unravel cIAP1/XIAP Molecular Inhibition Mechanisms

open access: yesAsian Journal of Organic Chemistry, Volume 15, Issue 7, July 2026.
The phosphoroorganic modification of Smac‐mimetics (SMs) drives selectivity for cIAP1‐ over XIAP‐BIR3. The divalent SM 6 partially dimerizes cIAP1, whereas it prefers intramolecular binding in XIAP. In XIAP the phosphoroorganic moiety of the monovalent SM 7 functions as an IBM, thus inhibiting both BIR2 and BIR3. Created in BioRender.
