SPHINX31 acts as a SRPK1 inhibitor targeting the ATR/DNA‐PKcs/CHK1 replicative checkpoint to inhibit cell growth in non‐small cell lung cancer

open access: yesMolecular Oncology, EarlyView.
The kinase SRPK1 directly interacts with the protein TOPBP1 and regulates the pre‐mRNA splicing of WIZ thereby contributing to the activation of the ATR/CHK1 replicative checkpoint in response to replicative stress. This allows cancer cells' genomic stability and survival.

Fragment-Based Discovery of an Apolipoprotein E4 (apoE4) Stabilizer

open access: yes, 2019
Apolipoprotein E is a 299-residue lipid carrier protein produced in both the liver and the brain. The protein has three major isoforms denoted apoE2, apoE3, and apoE4 which differ at positions 112 and 158 and which occur at different frequencies in the ...

Mutant p53R273H disrupts PDPK1 homodimerization and contributes to PDPK1 activation

open access: yesMolecular Oncology, EarlyView.
How mutant p53R273H drives AKT signaling is unclear. We show that p53R273H, but not wild‐type, directly binds PDPK1 via a mutation‐dependent conformational change. This interaction disrupts inhibitory PDPK1 homodimerization and enhances AKT phosphorylation.

NMR methods for fragment selection

open access: yes, 2014
International audienceFragment screening resides in the identification of minimal motifs responsible for the specific protein-ligand recognition. Once fragment hits have been identified, one needs to ensure that the fragments exhibit a specific binding ...

Analysis of movement laws of fragment and shock wave from a blast fragmentation warhead

open access: yesJournal of Measurement Science and Instrumentation, 2015
By studying shock wave propagation and fragment movement after the explosion of a blast fragmentation warhead,a whole calculation process about the place where fragments meet shock wave was proposed.A computing system for movement laws of fragments and ...

Targeting the EpCAM‐AXL axis to overcome drug resistance in lung cancer

open access: yesMolecular Oncology, EarlyView.
Lung cancer cells often evade therapy by hijacking signaling pathways. We reveal that cleaved EpCAM (sEpCAM) stabilizes the oncogenic protein AXL, driving NF‐κB and STAT3‐mediated chemoresistance. This EpCAM‐AXL axis identifies a high‐risk patient subset with poor prognosis.

Characterization of therapeutic antibodies targeting lymphatic and vascular resistance factor for the treatment of hypervascularized and highly metastatic cancers

open access: yesMolecular Oncology, EarlyView.
LVRF (lymphatic and vascular resistance factor) is a VEGFA splice variant upregulated in response to anti‐angiogenic therapy that fosters tumor adaptation through enhanced vascularization, lymphangiogenesis, and metastatic dissemination. Targeting LVRF with fully human neutralizing antibodies disrupts these processes and limits resistant tumor growth ...

The multifaceted roles of lysosomes in cancer

open access: yesMolecular Oncology, EarlyView.
Lysosomes support cancer cell adaptation by integrating macromolecule degradation and nutrient recycling with nutrient sensing, inter‐organelle communication and immune regulation. These interconnected functions can promote tumour growth, stress tolerance and therapy resistance, while also creating context‐dependent vulnerabilities that may be ...
