FRET-based peptide screening assay.
Cartoon representation of the conjugation of mRuby2-Q-tag and Clover-K-tag, in the presence of bTG, resulting in a cross-linked product. Due to the spectral overlap of mRuby2 and Clover, when in close proximity, excitation of Clover leads to FRET, and ...

The Shewanella oneidensis Fic enzyme SoFic targets the switch‐I region of EF‐Tu for AMPylation
Fic enzymes mediate diverse post‐translational modifications across all domains of life, including AMPylation. Prokaryotic EF‐Tu can be AMPylated and deAMPylated by the conserved Fic enzyme SoFic. Structural and biochemical approaches were used to characterize the effect of AMPylation on EF‐Tu, SoFic's enzymatic activities, and the enzyme‐target ...

FRET-Modulated Multihybrid Nanoparticles for Brightness-Equalized Single-Wavelength Barcoding
Semiconductor quantum dots (QDs) are the most versatile fluorophores for Förster resonance energy transfer (FRET) because they can function as both donors and acceptors for a multitude of fluorophores.

We identify USP29 as the only DUB mirroring CA9 expression, a marker of hypoxia and HIF pathway activation associated with PCA aggressiveness. USP29 stabilizes HIF‐1α and HIF‐2α via a noncanonical mechanism that is independent of PHD/pVHL activity yet relies on proteasomal regulation, establishing USP29 as a previously unrecognized regulator of hypoxic

FRET-TnC constructs varying the FRET pair of fluorophores and linker length.
Five constructs were generated by flanking human cardiac troponin C (cTnC) with one of three FRET pairs of fluorophores. Constructs also differ by the linker length (indicated by number of amino acids, aa) separating the fluorescent proteins from cTnC.

Reprogramming tumor cells with LCOR mRNA nanotherapy to enhance immunotherapy
LCOR (Ligand‐dependent corepressor) mRNA encapsulated in polymeric nanoparticles restores LCOR activity in tumor cells, constraining aggressive tumor cell states and increasing tumor immunogenicity. This reprogramming makes tumor cells more vulnerable to T‐cell attack and enhances their response to immune‐checkpoint immunotherapy.
