FRETting Mice Shed Light on Cardiac Adrenergic Signaling [PDF]
See related article, pages 1084–1091 β-adrenergic receptor (β-AR) signaling in cardiac myocytes influences contractile and relaxation states in the heart. Classically, following hormone activation, β-AR preferentially couples with Gs, which in turn activates adenylyl cyclase and cAMP production.

Pediatric Idiopathic Multicentric Castleman Disease Is Often Severe But Responsive to Siltuximab
ABSTRACT Background Idiopathic multicentric Castleman disease (iMCD) is a potentially fatal immunologic disorder marked by widespread lymphadenopathy and inflammation. Siltuximab, an interleukin‐6 (IL‐6) inhibitor, is the only FDA‐approved treatment for adult patients with iMCD.

Quantitative Visualization of Intracellular Nanometabolism Using Peak-Shifted Dual-State Emissive FRET Nanoprobes. [PDF]
This study reveals the long‐elusive intracellular dissolution mechanism of carrier‐free nanomedicines. By utilizing peak‐shifted dual‐state emissive FRET nanoprobes (PDFNPs) that undergo a distinct ratiometric fluorescence peak shift upon disassembly, we achieve real‐time, quantitative tracking of dissolution kinetics in live cells.

Investigating cardiac β-adrenergic nuclear signaling with FRET-based biosensors
By activating membrane β-adrenergic receptors (β-AR), noradrenaline and adrenaline are the most powerful stimulators of cardiac function. β-ARs are coupled to the synthesis of cAMP, which activates the cAMP-dependent protein kinase (PKA). PKA regulates the key proteins of excitation-contraction coupling but also gene expression.

We present robust protocols for the preparation of supported lipid bilayers (SLBs) incorporating either Salmonella smooth LPS or outer membrane vesicles (OMVs). We use a combination of quartz crystal microbalance with dissipation (QCM‐D) and fluorescence microscopy to both characterize the SLBs of various compositions and to probe their interactions ...

Structure‐forward targeting of claudins with synthetic binders
Claudins form the paracellular barriers between epithelial and endothelial tissues at tight junctions and are targets for molecular binders with the goal of modulating barrier permeability. Claudin‐binding molecules are relevant in drug delivery or in altering claudin interactions with disease‐causing proteins.

The Rac-FRET Mouse Reveals Tight Spatiotemporal Control of Rac Activity in Primary Cells and Tissues
The small G protein family Rac has numerous regulators that integrate extracellular signals into tight spatiotemporal maps of its activity to promote specific cell morphologies and responses.

Autoinducer-Fluorophore Conjugates Enable FRET in LuxR Proteins in Vitro and in Cells [PDF]
Cell-to-cell signaling, or quorum sensing (QS), in Gram-negative bacteria is governed by small molecule signals (N-acyl L-homoserine lactones, AHLs) and their cognate intracellular receptors (LuxR-type proteins).

Synergistic perspectives—How single‐molecule biophysics complement biochemical understanding
In this review, we discuss how ensemble biochemistry and single‐molecule approaches are complementary, outline commonly used single‐molecule techniques, and illustrate their relevance through two representative case studies: chromatin organization by SMC complexes and pathway choice during DNA double‐strand break repair.
