Aberrant GALNT7‐mediated O‐GalNAcylation stabilizes TAZ to drive gallbladder cancer progression through a feed‐forward transcriptional loop. Structure‐based screening identifies Olaparib as a potent GALNT7 antagonist that disrupts this oncogenic axis, providing an immediate therapeutic strategy for this aggressive malignancy.

Cardiac G-Protein–Coupled Receptor Kinase 2 Ablation Induces a Novel Ca 2+ Handling Phenotype Resistant to Adverse Alterations and Remodeling After Myocardial Infarction [PDF]
G-protein-coupled receptor kinase 2 (GRK2) is a primary regulator of β-adrenergic signaling in the heart. G-protein-coupled receptor kinase 2 ablation impedes heart failure development, but elucidation of the cellular mechanisms has not been achieved ...

G Protein–Coupled Receptor Kinase 2 [PDF]
Heart failure (HF) causes a tremendous burden on the worldwide healthcare system, affecting >23 million people. There are many cardiovascular disorders that contribute to the development of HF and multiple risk factors that accelerate its occurrence, but regardless of its underlying cause, HF is characterized by a marked decrease in myocardial ...

With increasing knowledge of the regulatory mechanisms of G-protein-coupled receptor signaling in heart physiology, many studies have focused on the role of this system in cardiovascular disease. In recent years, scientists have moved their attention from the receptors to their regulatory proteins: the G-protein-coupled receptor kinases.

In this study, we investigated the regulation of different G protein-coupled receptor (GPCR)-stimulated signaling pathways by GPCR kinase 2 (GRK2). We used thyrotropin receptor, which is coupled to different G proteins, to investigate the regulation of ...

Regulation of G protein-coupled receptor kinase 2
2002Publisher Summary G protein-coupled receptor kinase 2 (GRK2) was originally termed a “β-adrenergic receptor kinase” (βARK) because it was purified as a kinase of the adrenergic β 2 receptor, and the adrenergic β 2 receptor was the only known substrate of GRK2. GRK2 is now known to phosphorylate different kinds of G protein-coupled receptors (GPCRs),
