FAK signaling drives chemoresistance and tumor evolution in pancreatic ductal adenocarcinoma. The FAK inhibitor IN10018 restores gemcitabine sensitivity by inducing SLC7A11‐mediated ferroptosis through the PI3K‐Akt pathway and remodeling the tumor microenvironment, reducing mesenchymal components while enhancing CD8+ T‐cell infiltration in preclinical ...

Dual priming with minocycline and photodynamic priming reprograms the pancreatic tumor microenvironment to overcome chemoresistance. By suppressing DNA repair enzyme Tdp1, inducing photooxidative damage, and enhancing irinotecan delivery via light‐activated nanoencapsulation, this strategy remodels the tumor microenvironment and drives immune ...

Elevated PAF1-RAD52 axis confers chemoresistance to human cancers
Summary: Cisplatin- and gemcitabine-based chemotherapeutics represent a mainstay of cancer therapy for most solid tumors; however, resistance limits their curative potential.

Nucleic Acid Therapeutics for “Undruggable” Cancer Targets: Mechanisms, Challenges, and Prospects
Nucleic acid therapeutics bypass the structural limitations of conventional drugs by targeting mRNA rather than proteins. This review examines how antisense oligonucleotides, siRNAs, miRNAs, aptamers, and mRNA vaccines intervene against historically undruggable oncoproteins including Ras, MYC, and p53, highlighting mechanistic advances, delivery ...

This non‑enzymatic function of DHODH drives sunitinib resistance by competing with TRIM37 to block TRIM28 ubiquitination, thereby stabilizing TRIM28 and activating VEGFA transcription. Disrupting the DHODH–TRIM28 interaction with lisaftoclax restores drug sensitivity.

Background Lactate dehydrogenase A (LDHA) can regulate tumorigenesis and cancer progression. Nevertheless, whether the regulation of LDHA is involved in the development of gemcitabine resistance in PDAC has not yet been fully elucidated.

Clinical application and drug resistance mechanism of gemcitabine
Gemcitabine, as a nucleoside analog, exerts a broad-spectrum antitumor effect by interfering with DNA synthesis, but its clinical application is limited by drug resistance. The drug resistance mechanism involves metabolic abnormalities (such as downregulation of deoxycytidine kinase (dCK), nucleoside transporter hENT1 deficiency), enhanced DNA repair ...

ABSTRACT Mosunetuzumab plus polatuzumab vedotin has shown promising activity versus rituximab plus polatuzumab vedotin (R‐Pola) in patients with relapsed/refractory (R/R) large B‐cell lymphoma (LBCL; NCT03671018). We present results from the Phase II randomized cohort, evaluating subcutaneous mosunetuzumab plus polatuzumab vedotin (Mosun‐Pola), with ...
