Hyperandrogenemia Induces Trophoblast Ferroptosis and Early Pregnancy Loss in Patients With PCOS via CMA‐Dependent FTH1 Degradation

open access: yesAdvanced Science, EarlyView.
In PCOS patients with hyperandrogenemia, decreased ferritin heavy chain 1 (FTH1) causes Fe2⁺ overload and ferroptosis in trophoblasts. Androgens induce FTH1 protein degradation via AR‐LAMP2A‐mediated chaperone‐mediated autophagy pathway, leading to placental development disruption and early pregnancy loss. Metformin mitigates androgen‐induced placental

Dynamics of the Mammalian Placental Metabolome in Placentogenesis and Embryonic Development

open access: yesAdvanced Science, EarlyView.
This study identifies three metabolic stages (E8.5, E9.5–10.5, E11.5–14.5) and two transition periods (E8.5–9.5, E10.5–11.5) in mouse placental development. NAD(H) emerges as a key dynamic metabolite that enhances embryonic growth through accelerated segmentation and increased proliferation of mouse embryonic stem cell (mESC)‐induced presomitic ...

Tim1 Deficiency Mediates Gestational Hyperglycemia‐Related Syncytiotrophoblast Dysfunction and Fetal Growth Restriction

open access: yesAdvanced Science, EarlyView.
Hyperglycemia during pregnancy impairs the fusion of trophoblast cells into syncytiotrophoblasts, leading to fetal growth restriction. This impaired fusion is mediated by Tim1 downregulation via hyperglycemia‐induced ROS. Antioxidant therapy during pregnancy promotes syncytiotrophoblast formation by upregulating Tim1 expression, thus alleviating fetal ...

Comparing and Evaluating Five-Day Chemotherapy Agents Actinomycin D and Methotrexate in Low-Risk Post-Molar Gestational Trophoblastic Neoplasia: A Retrospective Analysis

open access: yesClinical and Experimental Obstetrics & Gynecology, 2022
Background: Low-risk post-molar gestational trophoblastic neoplasia is sensitive to chemotherapy, but there is no consensus regarding the best treatment regimen for low-risk post-molar gestational trophoblastic neoplasia.

Decoding Human Placental Cellular and Molecular Responses to Obesity and Fetal Growth

open access: yesAdvanced Science, EarlyView.
Women with obesity often deliver large‐for‐gestational‐age (LGA) infants. Single‐nucleus RNA sequencing of term placenta reveals that hypoxia and TNF‐α signaling in syncytiotrophoblasts are featured in maternal obesity, but inflammatory signatures in Hofbauer cells and response to lipid or carbohydrate metabolism in fibroblasts are specific to LGA.

The role of surgery in the management of women with gestational trophoblastic disease

open access: yesRevista do Colégio Brasileiro de Cirurgiões
The Gestational Trophoblastic Disease includes an interrelated group of diseases originating from placental tissue, with distinct behaviors concerning local invasion and metastasis.

Downregulation of the gli transcription factors regulator Kif7 facilitates cell survival and migration of choriocarcinoma cells. [PDF]

open access: yesPLoS ONE, 2014
The kinesin protein Kif7 has been recognized as an integral component of hedgehog signalling. Aberrant activation of hedgehog signalling has been implicated in many human solid tumours.

HRS Degradation‐Induced Nicotinamide Deficiency in Placental Extracellular Vesicles Triggers Preeclampsia by Disrupting Maternal‐Fetal Immune Homeostasis

open access: yesAdvanced Science, EarlyView.
This study shows that lower NAM levels in PE‐derived pEVs correlate with disease severity. NAM‐deficient pEVs reduce Th1 and Th17 inhibition, leading to PE‐like symptoms. NAM in pEVs inhibits Th1 via SIRT1 and Th17 via macrophages. Reduced NAM in PE‐EVs is due to decreased HRS expression in trophoblasts, resulting from elevated HSP27.
