Prognostic Factors of Renal Involvement in Systemic Sclerosis [PDF]

open access: yes, 2018
Background/Aims: Renal involvement is common in systemic sclerosis (SSc), including asymptomatic reduction of glomerular filtration rate (GFR), increased renal resistance indices, scleroderma renal crisis (SRC) and ANCA-associated vasculitis. The aim was

Translation of a physiologically‐based pharmacokinetic model for dabigatran etexilate to the design of a safety and efficacy study in post‐partum women

open access: yesBritish Journal of Clinical Pharmacology, EarlyView.
Aims To translate a PBPK model developed for the direct oral anticoagulant, dabigatran etexilate, the prodrug of dabigatran, based on data obtained from healthy men to healthy non‐pregnant, pregnant and post‐partum women. To evaluate safety and efficacy of dabigatran etexilate in post‐partum women using simulations and design a future clinical study to

Measurement of Glomerular Filtration Rate Using Multiphasic Computed Tomography in Patients With Unilateral Renal Tumors: A Feasibility Study

open access: yesFrontiers in Physiology, 2019
ObjectivesThis study was to assess the feasibility of a modified multiphasic CT scan protocol combined with homemade software measurements of glomerular filtration rate (CT-GFR) and explore the effect of renal tumor volume on the calculation of CT-GFR ...

Measured GFR by Utilizing Population Pharmacokinetic Methods to Determine Iohexol Clearance

open access: yesKidney International Reports, 2020
Introduction: There is an increasing demand for accurately measured glomerular filtration rate (GFR). Iohexol serum clearance has become a new gold standard, but it is challenging when GFR is low and 24-hour sampling is required for accurate results. The

Renal function assessment in adults with recurrent calcium kidney stone disease [PDF]

open access: yes, 2012
The prevalence of nephrolihiasis and chronic kidney disease has risen over the past three decades, we sought to determine if person with a history of kidney stones have lower renal function relative to non stone formers.

A real‐world data‐driven approach to optimizing enoxaparin dosing in burn patients

open access: yesBritish Journal of Clinical Pharmacology, EarlyView.
Aim Enoxaparin dosing in burn patients is challenging due to physiological changes affecting absorption, distribution and clearance. The objectives of this study are to develop a population pharmacokinetic (PK) model for enoxaparin in burn patients, evaluate target attainment with current equation‐based (EQ) dosing and propose an optimized dosing ...

Effects of Renal and Hepatic Impairment on the Pharmacokinetics of Zilurgisertib

open access: yesBritish Journal of Clinical Pharmacology, EarlyView.
Aim Small‐molecule activin receptor‐like kinase‐2 inhibitor zilurgisertib (INCB000928) is under investigation in a pivotal trial for the treatment of fibrodysplasia ossificans progressiva (FOP), an ultrarare genetic condition. This analysis assessed effects of renal and hepatic impairment on zilurgisertib pharmacokinetics.

Simultaneous measurement of kidney function by dynamic contrast enhanced MRI and FITC-sinistrin clearance in rats at 3 tesla: initial results.

open access: yesPLoS ONE, 2013
Glomerular filtration rate (GFR) is an essential parameter of kidney function which can be measured by dynamic contrast enhanced magnetic resonance imaging (MRI-GFR) and transcutaneous approaches based on fluorescent tracer molecules (optical-GFR). In an

Blood pressure effects of SGLT2 inhibitors and GLP‐1 receptor agonists: Mechanisms, trial evidence and Real‐world data

open access: yesBritish Journal of Clinical Pharmacology, EarlyView.
SGLT2 inhibitors and GLP‐1 receptor agonists modestly lower blood pressure across diverse patient populations, including those without diabetes. These effects appear largely independent of glycaemic control and offer additive value in high‐risk patients with overlapping comorbidities.
