Expanding Genetic Code to Generate Human Brain Organoids with Both Vasculature and Microglia‐Like Cells

Using genetic code expansion, we engineered vascularized human cerebral organoids (vhCOs) with microglia‐like cells and blood‐brain barrier features. vhCOs recapitulate neurovascular interactions, regional identities, and neuronal subtypes resembling the fetal brain.

The GPX3‐VCAM1 Axis Gates Pro‐Fibrotic Tubule Cell Fate in Hyperuricemic Nephropathy

Hyperuricemia induces NLRP3 inflammasome signaling activation and downregulation of GPX3 in proximal tubular epithelial cells, leading to oxidative stress–driven VCAM1 upregulation, emergence of pro‐fibrotic proximal tubule cells, macrophage recruitment, and renal fibrosis.

Microglial TSPAN4‐Dependent Migrasomes Promote Pathological Retinal Neovascularization via Immune‐Vascular Crosstalk

Combined high‐glucose and hypoxic stress switches microglia into a TSPAN4‐dependent migrasome‐producing state. These migrasomes deliver pro‐angiogenic signals to endothelial cells, activating HIF‐1α/VEGF signaling, disrupting vascular junctions, and fueling pathological neovascularization. Spatiotemporally Controlled Sequential Chemo‐Immunotherapy Activates Pyroptosis for Robust Anti‐Tumor Immunotherapy

A glutathione and ultrasound dual‐responsive cerasome achieves sequential co‐delivery of gemcitabine and BMS1166 in tumors. Chemotherapy priming followed by on‐demand PD‐L1 blockade synergistically amplifies anti‐tumor immunity, inhibiting primary tumor growth, distant metastasis and inducing durable immune memory.
