Synthesis and biological evaluation of glaucocalyxin A derivatives as potential anticancer agents

European Journal of Medicinal Chemistry, 2014
A series of Mannich base type derivatives of Glaucocalyxin A (GLA) were designed and prepared. The cytotoxicity of these compounds was evaluated against six tumor cell lines (SMMC-7721, B16, SGC-7901, A549, KB, HL-60). Most compounds exhibited potent antiproliferative effects with low micromolar IC50 values.

Glaucocalyxin A improves survival in bleomycin-induced pulmonary fibrosis in mice

Biochemical and Biophysical Research Communications, 2017
Idiopathic pulmonary fibrosis (IPF) is a progressive interstitial lung disease with unclear etiology and poor prognosis. Despite numerous studies on the pathogenesis of IPF, only scant treatment options are available for the management of IPF. Glaucocalyxin A (GLA), an ent-Kaurane diterpenoid, has been demonstrated to exert anti-inflammatory, anti ...

Simultaneous determination of glaucocalyxin A and glaucocalyxin B in rat plasma by LC‐MS/MS and its application to a pharmacokinetic study after oral administration of Rabdosia japonica extract

Biomedical Chromatography, 2017
AbstractA specific and sensitive liquid chromatography tandem mass spectrometry (LC‐MS/MS) method was developed and validated for the analysis of glaucocalyxin A and glaucocalyxin B in rat plasma using praeruptorin A as an internal standard. Separation was performed on a Hypurity C18 column (2.1 × 50 mm, 5 μm) with isocratic elution using 0.2% formic ...

Glaucocalyxin A induces apoptosis and autophagy in tongue squamous cell carcinoma cells by regulating ROS

Cancer Chemotherapy and Pharmacology, 2021
Tongue squamous cell carcinoma (TSCC) is the most common highly invasive oral cancer. Glaucocalyxin A (GLA) is a diterpenoid component isolated from Rabdosia japonica var. with anti-bacterial and anti-cancer biological properties. However, the role of GLA in human TSCC remains uncertain.

Synthesis and Cytotoxicity Assessment of Novel 7-O- and 14-O-Derivatives of Glaucocalyxin A

Anti-Cancer Agents in Medicinal Chemistry, 2020
Background: Rabdosia japonica has been historically used in China as a popular folk medicine for the treatment of cancer, hepatitis, and gastricism. Glaucocalyxin A (GLA), an ent-kaurene diterpene isolated from Rabdosia japonica, is one of the main active ingredients showing potent inhibitory effects against several types of tumor cells.

Glaucocalyxin A attenuates angiotensin II-induced cardiac fibrosis in cardiac fibroblasts

Biochemical and Biophysical Research Communications, 2018
Glaucocalyxin A (GLA) is a natural ent-Kaurane diterpenoid that possesses cardioprotective effect. Recently, it has been reported that GLA inhibits liver and pulmonary fibrosis, whereas its role in cardiac fibrosis remains unknown. In the present study, we evaluated the effect of GLA on the pathogenesis of cardiac fibrosis in vitro.

Chemoproteomics Reveals Glaucocalyxin A Induces Mitochondria‐Dependent Apoptosis of Leukemia Cells via Covalently Binding to VDAC1

Advanced Biology, 2023
AbstractChronic myelogenous leukemia (CML) that is resistant to tyrosine kinase inhibitors is one of the deadliest hematologic malignancies, and the T315I mutation in the breakpoint cluster region‐Abelson (BCR‐ABL) kinase domain is the most prominent point mutation responsible for imatinib resistance in CML.
