Consequences and Mechanisms of Noise‐Induced Cochlear Synaptopathy and Hidden Hearing Loss, With Focuses on Signal Perception in Noise and Temporal Processing

open access: yesAdvanced Science, EarlyView.
Noise‐induced synaptopathy (NIS) is largely reversible due to self‐repair. NIS and noise‐induced hidden hearing loss are two concepts with similarities and differences. The major hearing deficits in NIHHL are temporal processing disorders. The translation of animal data in NIS studies to humans is hindered by many factors.

Alnustone Ameliorates Metabolic Dysfunction‐Associated Steatotic Liver Disease by Facilitating Mitochondrial Fatty Acid β‐Oxidation via Targeting Calmodulin

open access: yesAdvanced Science, EarlyView.
This study identifies alnustone, a natural compound from Alpinia katsumadai, as a potent therapeutic agent for MASLD and MASH. Alnustone enhances mitochondrial fatty acid β‐oxidation by directly targeting calmodulin, improving liver steatosis, fibrosis, and insulin resistance in vivo.

Human Bone‐Derived Endothelial Cells Mediate Bone Regeneration via Distinct Expression of KIT Ligand

open access: yesAdvanced Science, EarlyView.
Bone‐specific endothelial cells (b‐ECs) uniquely express KITLG, which recruits c‐Kit+/CD34+ hematopoietic progenitors (HPCs) to the osteovascular niche, initiating a cascade that culminates in the osteogenic differentiation of bone marrow‐derived mesenchymal stem cells (bm‐MSCs) and subsequent ossification.

The Nuclear Localization of ACLY Guards Early Embryo Development Through Recruiting P300 and HAT1 to Promote Histone Acetylation and Transcription

open access: yesAdvanced Science, EarlyView.
ACLY is vital for early embryo development. IGF‐1 activates AKT to phosphorylate ACLY, driving its nuclear localization and recruitment of HATs (P300/HAT1), boosting acetyl‐CoA production and histone acetylation for transcriptional activation. Conversely, ACLY deficiency (via knockdown, knockout, or AKT inhibition) reduces nuclear acetyl‐CoA, disrupts ...

Chronic Psychological Stress Induces Cardiomyocyte Hypertrophy Through Corticosterone‐Glucocorticoid Receptor‐LAMA5 Axis

open access: yesAdvanced Science, EarlyView.
In vivo experiments suggest that cardiomyocyte hypertrophy is an early manifestation of cardiac pathological changes induced by chronic psychological stress. In vivo and in vitro experiments suggest that the corticosterone‐glucocorticoid receptor‐LAMA5 axis mediates cardiomyocyte hypertrophy induced by chronic psychological stress. The clinical results

PSMD14 Stabilizes SLC7A11 to Ameliorate Glucocorticoid‐Induced Osteoporosis by Suppressing Osteocyte Ferroptosis

open access: yesAdvanced Science, EarlyView.
Glucocorticoid‐induced osteoporosis (GIOP) triggers osteocyte ferroptosis via SLC7A11 degradation. PSMD14 stabilizes SLC7A11 by counteracting glucocorticoid‐driven ubiquitination, preserving cystine uptake and glutathione synthesis. AAV‐mediated PSMD14 delivery or its agonist Pantethine rescues osteocyte survival and bone loss in GIOP mice.
