A CRISPR/Cas12a‐powered dendritic DNA nanostructure is assembled to create confined enzymatic microdomains, enabling ultrasensitive detection. Here, we present a tetrahedral DNA framework (TDF)‐mediated Cas12a ordered assembly strategy. TDFs arrange CRISPR/Cas12a into ordered structures controllably, enhancing catalytic efficiency through localized Cas12a hotspots while maintaining spatial intervals to facilitate substrate diffusion.

This study developed a pair of duplex RPA‐LFA assays for the simultaneous detection of critical resistance genes, blaCTX‐M/mcr‐1 and blaoxa‐48‐like/blaNDM, from hemoculture samples. Under optimized conditions, duplex RPA was performed at 37°C–38°C for 25 min, followed by LFA for 10–15 min.
