S49 cyc- lymphoma cells contain a mutation resulting in loss of a functional guanine nucleotide regulatory protein rendering their adenylate cyclase refractory to most stimuli. S49 wild-type and cyc- clones were used in the present study to investigate the possible association of altered cAMP metabolism with tumorigenicity and metastatic potential. The

Postmortem cerebral cortex Gs α-subunit levels are elevated in bipolar affective disorder
Brain Research, 1991We examined the relative abundance of G-protein subunits in postmortem brain obtained from 7 patients with bipolar affective disorder (BAD) compared with 7 age- and sex-matched controls. G-protein subunit immunoreactivities were determined in membranes prepared from postmortem prefrontal cortex using SDS-PAGE and immunoblotting with specific polyclonal

Analysis of mutations in the Gs protein α subunit gene in human leukaemia
Leukemia Research, 1992Mutations at codons 201 and 227 that cause oncogenic activation of the gene encoding the Gs alpha subunit protein occur in a subset of human solid tumours. To determine whether such oncogenic mutations occur in leukaemia we have analysed DNA from leukaemic cells from 59 patients representing a spectrum of acute and chronic leukaemia types by direct ...

Overexpression of Gs alpha subunit in thyroid tumors bearing a mutated Gs alpha gene.
Journal of cancer research and clinical oncology, 1995Point mutations occurring at codon 201 of the gene coding for the alpha subunit of the stimulatory G protein impair intrinsic GTPase activity and lead to a constitutive activation of adenylate cyclase. We have examined thyroid follicular and papillary carcinomas and follicular adenomas and found samples that bear this mutation at codon 201 of the Gs ...

The effect of iloprost on the ADP-ribosylation of Gs alpha (the alpha-subunit of Gs).
The Biochemical journal, 1989Treatment of platelets with a prostacyclin analogue, iloprost, decreased the cholera-toxin-induced ADP-ribosylation of membrane-bound Gs alpha (alpha-subunit of G-protein that stimulates adenylate cyclase; 42 kDa protein) and a cytosolic substrate (44 kDa protein) [Molina y Vedia, Reep & Lapetina (1988) Proc. Fully-differentiated mouse 3T3-L1 fibroblasts accumulate large amounts of lipid at 7-10 days after induction by insulin or by dexamethasone and a methyl xanthine. G proteins mediate transmembrane signalling from a diverse group of cell-surface receptors to effector units that include phospholipase C, adenylyl cyclase and ion channels.

Lithium decreases Gs, Gi-1 and Gi-2 α-subunit mRNA levels in rat cortex
European Journal of Pharmacology: Molecular Pharmacology, 1991Male Wistar rats (250-275 g) were maintained on powdered rat chow containing 0.2% LiCl for 21 days; control animals were fed a similar diet without the added lithium. Animals were killed by decapitation and cerebral cortex was dissected, forzen and stored at −70 o C.

Trimeric G Protein α Subunits of the Gs and Gi Families Localized at the Golgi Membrane
Biochemical and Biophysical Research Communications, 1995The occurrence of individual G proteins within Golgi membranes from liver and from adrenal medulla were analyzed by Western blotting. Two splice variants of G alpha s and also G alpha i-3 were found in both tissues. Additionally, G alpha i-2, its 43-kDa splice variant and G alpha o were strongly labeled in Golgi of adrenal medulla.

Reciprocal regulation of Gs alpha by palmitate and the beta gamma subunit.
Proceedings of the National Academy of Sciences of the United States of America, 1997Hormonal activation of Gs, the stimulatory regulator of adenylyl cyclase, promotes dissociation of alpha s from G beta gamma, accelerates removal of covalently attached palmitate from the G alpha subunit, and triggers release of a fraction of alpha s from the plasma membrane into the cytosol. To elucidate relations among these three events, we assessed

GTP-binding proteins are key elements in coupling receptors to various effector systems. Using ADP-ribosylation by cholera (CTX) and pertussis (PTX) toxins and an immunodetection technique, we investigated the G protein expression profile in smooth muscle of stem villi vessels obtained from human term placentae.
