Parthenogenetic Activation of Porcine Oocytes with Guanosine-5′-O-(3′-Thiotriphosphate)1 [PDF]

open access: closedBiology of Reproduction, 1995
A possible mechanism by which sperm activate oocytes may involve a G protein-mediated signal-transducing pathway. Artificial stimulation of G proteins leads to events very similar to those at fertilization in oocytes of several species. Here we report the effects of activation of a G protein with guanosine-5'-O-(3'-thiotriphosphate) (GTP-gamma-S), a ...

Inositol 1,4,5-trisphosphate- and guanosine 5'-O-(3-thiotriphosphate)-induced Ca2+ release in cultured airway smooth muscle. [PDF]

open access: yesBr J Pharmacol, 1991
The interaction between inositol 1,4,5‐trisphosphate (InsP3) and guanosine 5′‐O‐(3‐thio triphosphate) (GTPγS) releasable calcium (Ca2+) pools was examined using 45Ca effluxes in permeabilized cultured airway smooth muscle cells from rabbit trachea. Addition of InsP3 or GTPγS caused a concentration‐dependent release of intracellular Ca2+.

Carbachol in the presence of guanosine 5′-O-(3-thiotriphosphate) stimulates the breakdown of exogenous phosphatidylinositol 4,5-bisphosphate, phosphatidylinositol 4-phosphate, and phosphatidylinositol by rat brain membranes

open access: hybridJournal of Biological Chemistry, 1989
The breakdown of exogenously added [3H]inositol-labeled phosphoinositides by rat brain cortical membranes was stimulated by the muscarinic cholinergic agonist carbachol. The stimulation required the presence of guanine nucleotide. Optimal conditions were similar to those described for guanosine 5'-O-(3-thiotriphosphate) (GTP gamma S) + carbachol ...

Guanosine 5'-O-(3-thiotriphosphate) as an analog of GTP in protein biosynthesis. The effects of temperature and polycations on the accuracy of initial recognition of aminoacyl-tRNA ternary complexes by ribosomes.

open access: hybridJournal of Biological Chemistry, 1986
Guanosine 5'-O-(3-thio)triphosphate (GTP gamma S) is a good analog of GTP in the reactions leading to the formation of a peptide bond in protein biosynthesis. It forms binary and ternary complexes with elongation factor Tu (EF-Tu), and with EF-Tu and aminoacyl-tRNA (aa-tRNA). In addition, it stimulates aa-tRNA binding to ribosomes. Although GTP gamma S

Phospholipase D activation in a cell-free system from human neutrophils by phorbol 12-myristate 13-acetate and guanosine 5'-O-(3-thiotriphosphate). Activation is calcium dependent and requires protein factors in both the plasma membrane and cytosol.

open access: hybridJournal of Biological Chemistry, 1991
Receptor-linked activation of phospholipase D has been demonstrated recently in a variety of intact cell types including granulocytes, but little is known about the enzyme, its cofactor requirements, and regulation. Using [3H]alkyllysophosphatidylcholine to prelable an endogenous phosphatidylcholine substrate pool in conjunction with ...

Evidence that a beta-adrenergic receptor-associated guanine nucleotide regulatory protein conveys guanosine 5'-O-(3-thiotriphosphate)- dependent adenylate cyclase activity.

open access: hybridJournal of Biological Chemistry, 1981
The guanine nucleotide regulatory protein component (N) of the frog erythrocyte membrane adenylate cyclase system appears to form a stable complex with the beta-adrenergic receptor (R) in the presence of agonist (H). This agonist-promoted ternary complex HRN can be solubilized with Lubrol.

Guanosine 5'-O-(3-thiotriphosphate) and guanosine 5'-O-(2-thiodiphosphate) activate G proteins and potentiate fibroblast growth factor-induced DNA synthesis in hamster fibroblasts.

open access: hybridJournal of Biological Chemistry, 1990

Guanosine 5'-O-(3-thiotriphosphate) reduces ADP-ribosylation of the inhibitory guanine nucleotide-binding regulatory protein of adenylyl cyclase (Ni) by pertussis toxin without causing dissociation of the subunits of Ni. Evidence of existence of heterotrimeric pt+ and pt- conformations of Ni.

open access: hybridJournal of Biological Chemistry, 1987
The effect of the addition of guanosine 5'-O-(3-thiotriphosphate) (GTP gamma S), the GTP analog which activates the inhibitory guanine nucleotide-binding regulatory protein of adenylyl cyclase (Ni), on the pertussis toxin-mediated ADP-ribosylation reaction was studied in detail.

Muscarinic acetylcholine receptor‐stimulated binding of guanosine 5′‐O‐(3‐thiotriphosphate) to guanine‐nucleotide‐binding proteins in cardiac membranes [PDF]

open access: closedEuropean Journal of Biochemistry, 1989
Receptor‐regulated binding of the labeled GTP analog, guanosine 5′‐O‐(3‐thiotriphosphate) ([35S]GTP[S]), to guanine‐nucleotide‐binding proteins (G‐proteins) was studied in porcine atrial membranes enriched in muscarinic acetylcholine (mACh) receptors. Binding of [35S]GTP[S] to the membranes was not or only slightly affected by the cholinergic agonist ...

Guanosine 5´-O-(3-Thiotriphosphate) and Cations Regulate Melatonin Receptors, and Melatonin Inhibits Cyclic AMP Production in the Spinal Cord

open access: closedNeurosignals, 1997
Effects of guanosine 5'-O-(3-thiotriphosphate) (GTP gamma S) and cations on 2-[125I]iodomelatonin binding were investigated in membrane preparations of the chicken spinal cord. At concentrations of 10 and 50 mumol/l, GTP gamma S dose-dependently increased (p < 0.05) the equilibrium dissociation constant (Kd) and depressed (p < 0.05) the maximum number ...
