Guanosine-5'-O-(3-thiotriphosphate) modifies kinetics of voltage-dependent calcium current in chick sensory neurons. [PDF]
Internal perfusion with the G-protein activator guanosine-5'-O-(3-thiotriphosphate) (GTP-gamma S) mimics the effect of noradrenaline and dopamine on the voltage-dependent calcium current in chick dorsal root ganglion (DRG) cells. With 100 microM GTP-gamma S in the pipette, the current at +10 mV was depressed by approximately 50%, with a 10-fold ...

Intracellular application of guanosine-5'-O-(3-thiotriphosphate) induces exocytotic granule fusion in guinea pig eosinophils. [PDF]
The mechanism of eosinophil secretion was studied in guinea pig eosinophils by measuring release of hexosaminidase from cell suspensions (greater than 98% pure) permeabilized with streptolysin-O and by whole-cell patch-clamp capacitance measurements. It is shown that release of eosinophil granule components occurs by an exocytotic mechanism in which ...

Ligand-Independent GLUT4 Translocation Induced by Guanosine 5′-O-(3-Thiotriphosphate) Involves Tyrosine Phosphorylation* [PDF]
AbstractTo delineate the signaling pathway leading to glucose transport protein (GLUT4) translocation, we examined the effect of microinjection of the nonhydrolyzable GTP analog, guanosine 5′-O-(3-thiotriphosphate) (GTPγS), into 3T3-L1 adipocytes. Thirty minutes after the injection of 5 mm GTPγS, 40% of injected cells displayed surface GLUT4 staining ...

Biochemical Engineering Perspective on cGAS: From Enzyme Discovery to Potential Industrial Application. [PDF]
This review examines the structural and enzymatic properties of cyclic GMP‐AMP synthase, including substrate promiscuity, homologs, and engineered variants. In addition, it evaluates potential expression systems for large‐scale enzyme production and analyzes reaction and purification strategies for the synthesis of cyclic dinucleotides to connect ...

Inositol 1,4,5-trisphosphate- and guanosine 5'-O-(3-thiotriphosphate)-induced Ca2+ release in cultured airway smooth muscle. [PDF]
The interaction between inositol 1,4,5‐trisphosphate (InsP3) and guanosine 5′‐O‐(3‐thio triphosphate) (GTPγS) releasable calcium (Ca2+) pools was examined using 45Ca effluxes in permeabilized cultured airway smooth muscle cells from rabbit trachea. Addition of InsP3 or GTPγS caused a concentration‐dependent release of intracellular Ca2+.

Induction of neurite formation in PC12 cells by microinjection of proto-oncogenic Ha-ras protein preincubated with guanosine-5'-O-(3-thiotriphosphate). [PDF]
Rat pheochromocytoma (PC12) cells differentiate to neuronal cells in response to nerve growth factor. It has been shown that microinjection of oncogenic but not proto-oncogenic p21 protein induces morphological differentiation in PC12 cells (D. Bar-Sagi and J. R. Feramisco, Cell 42:841-848, 1985).

Parthenogenetic activation of porcine oocytes with guanosine-5'-O-(3'-thiotriphosphate). [PDF]
A possible mechanism by which sperm activate oocytes may involve a G protein-mediated signal-transducing pathway. Artificial stimulation of G proteins leads to events very similar to those at fertilization in oocytes of several species. Here we report the effects of activation of a G protein with guanosine-5'-O-(3'-thiotriphosphate) (GTP-gamma-S), a ...

Based on a simplified version of the morphinan scaffold, 3‐[3‐(phenalkylamino)cyclohexyl]phenol analogues were designed, synthesized and evaluated for their µ‐opioid receptor (MOR) antagonist activity in vitro and in silico. Docking studies indicate a peculiar combination of C‐1 stereochemistry and N‐substitutions as feasibly essential for MOR‐ligand ...
