Improved outcomes of acute lymphoblastic leukemia after allogeneic blood or marrow transplantation with high-dose post-transplantation cyclophosphamide in the era of more effective pre-transplant therapy

open access: yesHaematologica
The therapeutic landscape in ALL has changed dramatically over the last decade. Allogeneic blood or marrow transplantation (AlloBMT) has also evolved and remains an important option for consolidation.

Dimethyl fumarate combined with cisplatin at subcytotoxic doses sensitizes cervical cancer toward ferroptosis and apoptosis through GSH restriction and p53 (re)activation

open access: yesMolecular Oncology, EarlyView.
Dimethyl fumarate (DMF) reduces growth of HPV‐positive cervical cancer spheroids and induces ferroptosis in cervical cancer cells via blocking SLC7A11/Glutathione (GSH) axis. Combination of subcytotoxic doses of DMF and cisplatin (CDDP) further suppresses spheroid growth and drives cell death in 2D culture models.

IMPDH inhibition enhances cytarabine efficacy in SAMHD1‐expressing leukaemia cells via guanine nucleotide depletion

open access: yesMolecular Oncology, EarlyView.
Cytarabine is a key therapy for acute myeloid leukaemia (AML), but its efficacy is limited by the dNTPase SAMHD1, which hydrolyses its active metabolite. Screening nucleotide biosynthesis inhibitors revealed that IMPDH inhibitors selectively sensitise SAMHD1‐proficient AML cells to cytarabine.

Low Dose of IL‐2 Application for Graft‐Versus‐Host Disease Prophylaxis Following Haploidentical Stem Cell Transplantation

open access: yesMedComm
Graft‐versus‐host disease (GVHD) are still key obstacles of haploidentical transplantation. Interleukin‐2 (IL‐2) could promote natural killer (NK) cells and T‐regulatory cells (Tregs) cells expansion in vitro and in vivo.

Establishment of a humanized patient‐derived xenograft mouse model of high‐grade serous ovarian cancer for preclinical evaluation of combination immunotherapy

open access: yesMolecular Oncology, EarlyView.
We have established a humanized orthotopic patient‐derived xenograft (Hu‐oPDX) mouse model of high‐grade serous ovarian cancer (HGSOC) that recapitulates human tumor–immune interactions. Using combined anti‐PD‐L1/anti‐CD73 immunotherapy, we demonstrate the model's improved biological relevance and enhanced translational value for preclinical ...

Better Response and Prognosis of Venetoclax Plus Hypomethylating Agents Over Intensive Chemotherapy in Young Adults With Newly Diagnosed ASXL1‐Mutated Acute Myeloid Leukemia

open access: yesCancer Medicine
Background ASXL1 is one of the most frequently mutated genes in acute myeloid leukemia (AML) and retains adverse‐risk status in intensively treated cohorts according to 2022 European Leukemia Net (ELN) risk criteria.

Intensive chemotherapy vs venetoclax/hypomethylating agents for patients aged 60 to 75 years with favorable, NPM1-mutated AML

open access: yesBlood Neoplasia
: Patients with nucleophosmin 1 (NPM1)–mutated acute myeloid leukemia (AML) without a FLT3-internal tandem duplication mutation are considered to have favorable-risk disease that may be cured with intensive chemotherapy (7+3; IC) alone.

Differential expression of cancer‐related genes supports prediction of poor response to first‐line treatments in T‐ALL pediatric patients with high minimal residual disease

open access: yesMolecular Oncology, EarlyView.
In the present work, we have identified a transcriptional signature based on the differential expression of six genes (BCL2&MAST4, HSH2D&LAT2, METRN&PITPNM2) that would facilitate the early detection of T‐cell acute lymphoblastic leukemia (T‐ALL) patients prone to a poor treatment response and could be implemented at diagnosis, along with other risk ...

Increased Epstein‒Barr virus reactivation following prophylaxis for cytomegalovirus infection after haploidentical haematopoietic stem cell transplantation

open access: yesJournal of Hematology & Oncology
Letermovir (LTV) prophylaxis is effective in reducing the incidence of clinically significant cytomegalovirus (CMV) infection (cs CMVi) after allogeneic haematopoietic stem cell transplantation (allo-HSCT).

E2A selectively regulates TGF‐β–induced apoptosis in KRAS‐mutant non‐small cell lung cancer

open access: yesMolecular Oncology, EarlyView.
Ability to induce apoptosis by TGF‐β is frequently lost in advanced lung adenocarcinoma despite intact TGF‐β signaling. We identify E2A as a mutant KRAS–dependent mediator of resistance to TGF‐β–induced apoptosis. TGF‐β induces E2A via SMAD3 in mutant KRAS cells, and E2A silencing restores apoptosis and enhances radiation response in cell lines ...
