The IKZF1 N159S mutation is associated with poor outcome and a distinct molecular profile in adult patients with AML

open access: yesBritish Journal of Haematology, Volume 206, Issue 5, Page 1373-1379, May 2025.
Summary IKZF1 mutations are recurrent alterations in acute myeloid leukaemia (AML), and hotspot point mutation, N159S, has recently been associated with unique gene expression and adverse risk. To better understand the molecular and clinical associations of IKZF1 N159S‐mutated AML, we performed a pooled analysis of 4136 AML patients.

Iron trapping in macrophages reshapes the homeostasis of the haematopoietic system

open access: yesBritish Journal of Haematology, Volume 206, Issue 5, Page 1485-1496, May 2025.
Deletion of ferroportin (Fpn) in myeloid cells results in iron trapping in macrophages and reshapes whole haematopoiesis with ageing. Impaired local iron supply to bone marrow (BM) leads to anaemia and microcytaemia, unbalanced megakaryopoiesis versus erythropoiesis and reduced BM cells starting from haematopoietic stem cells.

Indirect presentation of mismatched human leukocyte antigen‐B associates with outcomes of cord blood transplantation

open access: yesBritish Journal of Haematology, Volume 206, Issue 5, Page 1406-1417, May 2025.
This study analysed 1498 single‐unit cord blood transplantation (CBT) cases (2000–2018) to evaluate the impact of mismatched human leukocyte antigen (HLA) immunogenicity. Results highlighted that Class I epitopes from recipient‐mismatched HLA‐B were associated with higher transplantation‐related mortality (TRM) and lower neutrophil engraftment ...

How low can we go? Comparison of liberal and restrictive red cell transfusion thresholds in paediatric allogeneic haematopoietic stem cell transplantation: A randomized multicentre feasibility trial

open access: yesBritish Journal of Haematology, EarlyView.
Optimal red blood cell transfusion thresholds for children with bone marrow failure are uncertain. Children may be able to safely tolerate lower haemoglobin thresholds, thereby reducing transfusions, which have risks. This study in paediatric allogeneic haematopoietic stem cell transplant aimed to assess feasibility of recruitment and protocol ...
