Immunization of woodchucks with recombinant hepatitis delta antigen does not protect against hepatitis delta virus infection

To assess the role of immunization against hepatitis delta antigen in the prevention of hepatitis delta virus infection, woodchuck carriers of woodchuck hepatitis virus were immunized with a 64 amino acid portion of hepatitis delta antigen from its N-terminal region.

AbstractThe aim of this study was to evaluate whether patients with chronic hepatitis delta virus (HDV) infection treated with alpha interferon and subsequent loss of hepatitis B surface antigen (HBsAg) eliminate HDV. HDV RNA was detected in 26 of 28 patients with chronic delta hepatitis using the polymerase chain reaction.

Hepatitis Delta Antigen and RNA Polymerase II

Replication and transcription of HDV proceed via RNA-dependent RNA synthesis. These reactions are thought to be catalyzed at least in part by host RNA polymerase II (RNAPII). Hepatitis delta antigen (HDAg), which is critical for these processes, was recently proposed to function as a transcription elongation factor for RNAPII.

The hepatitis delta virus (HDV) genome has only one open reading frame, which encodes the viral small delta antigen. After RNA editing, the same open reading frame is extended 19 amino acids at the carboxyl terminus and encodes the large delta antigen.
